小鼠肺肾两虚型肺癌模型的建立

Establishment of Mice Model of Lung Cancer with Lung and Kidney Deficiency

摘要: 目的 建立小鼠肺肾两虚型肺癌模型。方法 24只C57BL/6雄性小鼠随机分为正常组、Lewis肺癌细胞皮下移植瘤模型组(简称模型组)和肺肾两虚型Lewis肺癌细胞移植瘤组(简称复合模型组),每组8只。将40只同品系小鼠随机分为Lewis肺癌细胞皮下移植瘤模型组(简称模型组)、肺肾两虚型Lewis肺癌细胞移植瘤组(简称复合模型组),每组20只,以观察生存期。采用烟熏刺激法连续14 d建立肺气虚模型,采用皮下注射氢化可的松25 mg/kg,1次/日,连续给药14 d建立肾虚模型, 将Lewis肺癌细胞(2×106个)注射于小鼠右腋部皮下,建立Lewis肺癌细胞移植瘤模型。观察小鼠的一般状况、饲料消耗量、体质量、生存时间、去瘤体重与移植瘤重量、脾脏指数、胸腺指数、肺脏组织病理学改变,并检测各组小鼠胸腺、脾脏和移植瘤CD4+CD25+Treg细胞的含量、Foxp3 mRNA的表达水平等评价肺肾两虚型肺癌模型的情况。结果 与模型组比较,复合模型组小鼠出现活动减少、睡眠时间缩短、蜷曲、毛发脱落、摄食减少、体质量下降、呼吸浅快、小便量多等肺肾两虚证的表现; 其中位数生存时间(19 d)显著缩短,生存期范围离散程度较模型组小(P<0.01); 脾脏指数显著降低(P<0.01); 小鼠胸腺和脾脏CD4+CD25+Treg细胞的含量明显增多,Foxp3 mRNA的表达水平上调(P<0.01)。结论 建立了小鼠Lewis肺肾两虚型肺癌动物模型,为研究中医药抗肺癌提供科学的实验工具。

Abstract: Objective To establish a mice model of lung cancer with lung and kidney deficiency. Methods A total of 24 eight-week-old male C57BL/6 mice were randomly divided into normal group, Lewis lung cancer model group (model group)and Lewis lung cancer with lung and kidney deficiency group (combined group),with 8 mice in each group. Mice in normal group received no treatment, mice in model group were injected subcutaneously to the right axilla with Lewis lung cancer cells. The combined group were injected with Lewis lung cancer cells and hydrocortisione (25mg/kg, 1 times/day, × 14 days) and stimulated with cigarettes (2 times/days, × 14 days).The model of lung cancer with lung and kidney deficiency was determined by observing behaviors and physical signs of the mice, detecting the changes of feed consumption, body weight, survival time, tumor weight, spleen index, thymus index, lung index and histopathology. The changes of number of CD4+CD25+Foxp3+Treg cells and the expression of Foxp3 mRNA in spleen, thymus and tumor were detected by flow cytometer and real-time PCR. Results Compared with those in the normal group and model group, the mice in the combined group were noted with obvious signs of lung and kidney deficiency, such as activity less, sleepless, curly, hair loss, tiredness, obvious weight loss, rapid shallow breathing and urinating more than the amount of the performance, et al. Especially, the median survival time in combined group was significantly reduced 19 (13-33) d (P<0.01). The number of CD4+CD25+Foxp3+Treg cells in spleen and thymus was higher in combined group than in normal group and model group (P<0.01). The expression of Foxp3 mRNA in spleen and thymus was significantly up-regulated, compared with those in combined group than in normal group and model group (P<0.01). Conclusion The mice model of lung cancer with lung and kidney deficiency is established. It is provided an experimental tool for the study the mechanism Traditional Chinese Medicine treatment with lung cancer.
