改良慢性马兜铃酸肾病大鼠模型的建立

Establishing Modified Model of Chronic Aristolochic Acid Nephropathy In Rats

摘要: 目的 建立慢性马兜铃酸肾病(AAN)大鼠模型.方法 将雄性SD大鼠随机分为正常对照组、模型组,对照组灌服生理盐水,模型组每天灌服马兜铃酸单体5mg/kg,建立慢性AAN动物模型.各组分别在第2、4、6、8周留取24h尿液,在第4、8周取血,分离血清,检测肾功能指标,第8周留取肾组织,观察病理变化.结果 随着饲养时间的延长,马兜铃酸模型组尿β2-MG、尿NAG水平明显升高,肾小管间质病理损害明显.结论 改良方法建立的慢性AAN动物模型,对于药物改善由于马兜铃酸引起的慢性肾小管间质病变提供了新的实验对象.

Abstract: Objective To establish chronic aristolochic acid (AA) nephropathy in rats. Methods Male SD rats were randomly divided into control and model group. The model group rats were given daily with AA 5 mg/kg intragastically,the control group rats were given equal amount of normal saline. The 24h urine and blood samples were collected for detection of indexes of renal function, and at the end of the 8th week, take their nephridial tissue to test the change of pathology. Result Along with feeding times,the level of the urine β2-MG and NAG enzyme exertion increased markedly and the damage of tubular inters titium after giving AA in tset groups. Conclusion The model of chronic A AN in rat had been finished basically.
