高脂喂养大鼠常用胰岛素敏感性测定方法的评估

Evaluation of Methods to Detect Insulin Sensitivity in High-fat Diet Rats

摘要: 目的 探讨高脂喂养大鼠常用胰岛素敏感性测定方法与正糖高胰岛素钳夹技术的相关性。方法 30只雄性Wistar大鼠随机分为2组,正常对照组(NC, n=15)和高脂喂养组(HF, n=15),分别给予基础饲料和高脂饲料喂养,喂养10周后进行正糖高胰岛素钳夹试验、胰岛素耐量试验和葡萄糖耐量试验,进行大鼠胰岛素敏感性相关指标测定及指标间相关性分析。结果 HF组大鼠葡萄糖输注率[GIR (0.90±0.15 vs 4.97±0.68)mg·kg-1·min-1, P<0.01]和胰岛素耐量试验血糖自然对数的下降斜率KITT(24.41±6.77 vs 48.61±4.71, P<0.01)均显著低于NC组,且KITT与GIR呈显著相关(r=0.911, P<0.01)。结论 胰岛素耐量试验与正糖高胰岛素钳夹试验相关性显著。

Abstract: Objective To compare several frequently used surrogate indexes of insulin sensitivity and the euglycemic-hyperinsulinemic clamp experiments in high-fat diet rats. Methods Thirty male Wistar rats were randomly divided into two groups with a regular (NC group, n=15) or high-fat diet (HF group, n=15) for 10 weeks.The euglycemic-hyperinsulinemic clamp experiments,the insulin tolerance tests as well as the glucose tolerance tests were performed and the correlations of the euglycemic-hyperinsulinemic clamp experiments with different types of IR indexes were conducted. Result The glucose infusion rate [GIR (0.90±0.15 vs 4.97±0.68)mg·kg-1 ·min-1, P<0.01and KITT (24.41±6.77 vs 48.61±4.71, P<0.01) in HF group decreased significantly. The insulin tolerance tests correlated with the euglycemic-hyperinsulinemic clamp experiments significantly (r=0.911, P<0.01). Conclusion This study demonstrates that the insulin tolerance tests correlated with the euglycemic-hyperinsulinemic clamp experiments significantly.
