Effects of Hypoxic Training on Blood EPO, Hb and RBC Level in Rats

摘要: 目的 观察为期7 d的低氧训练对大鼠血液内促红细胞生成素(Erythropoietin,EPO)水平、血红蛋白(Hemoglobin, Hb)水平和红细胞(Red blood cell, RBC)数量的影响,以获得大鼠对低氧训练产生适应的实验证据及为建立适宜的低氧训练模式提供思路。方法 将动物置于不同低氧浓度环境2 h/d,大鼠跑台运动时间为30 min/d。使用ELISA方法检测大鼠血清中EPO含量; 采用全血自动分析仪检测血液中RBC数量和Hb水平。结果 (1)在11%和15%氧浓度下暴露2 h/d,大鼠血液内EPO未产生明显变化,但在15%氧浓度下暴露的同时进行运动,可明显提高大鼠血液内EPO含量; (2)无论是在11%和15%氧浓度下暴露还是在11%和15%氧浓度下运动,均可明显提高大鼠血液内RBC数量; (3)在11%和15%氧浓度下暴露可明显提高大鼠血液中Hb水平; (4)低氧训练结束15 d后,大鼠血液内RBC和Hb水平恢复至低氧训练前水平。结论 2 h/d、持续7 d的低氧刺激,可明显诱导SD大鼠RBC和Hb生成,但持续时间不足15 d。

Abstract: Objective To investigate effects of 7 day hypoxic training on the level of Erythropoietin(EPO), hemoglobin(Hb) and red blood cell (RBC) in blood of SD rats, and to obtain the experimental evidence of adaptation to hypoxic training in rats. Methods The rats were exposed in different concentration of hypoxic environment for 2h/day, the time of treadmill running of rats is 30min/day. The level of EPO was checked by ELISA, and the level of RBC and Hb were measured by automatic blood cell analyzer. Results (1) No effects of hypoxic exposure in 15% O2 and 11% O2 environment on EPO level were found significantly, but treadmill running in 15% O2 environment enhance EPO level in blood of rats. (2) The number of RBC was increased significantly in rats which exercised on treadmill or stayed in 15% O2 or 11% O2 environment. (3) The increase of concentration of Hb was found significantly in rats which stayed in 15% O2 or 11% O2 environment. (4) Effects of hypoxic training on RBC and Hb level disappear in rats 15days after hypoxic training. Conclusion Production of RBC and Hb are significantly induced by hypoxic training in rats, but lasting time of enhancing effect is shorter than 15 days.
