大鼠内耳组织微量核酸的提取及PCR扩增

Isolation and PCR Amplification of Nucleic Acid from Rat's Inner Ear

摘要: 分别用提质粒法,SDS-蛋白酶K法,TRIZOL法提取大鼠内耳组织微量核酸,对所提核酸进行紫外吸收光谱分析及PCR扩增。结果表明,提质粒法可制备不含核DNA的线粒体DNA;SDS-蛋白酶K法可制备总DNA;TRIZOL法可同时制备总DNA和总RNA。

Abstract: Three methods were used to isolate nucleic acid from rat’s inner ear. The amount and the pureness of the isolated nucleic acid were analyzed by PCR and UV-spectrometer. The results indicate that the three methods all can prepare high-quality nucleic acid from rat’s inner ear:method of plasmid preparation is good for isolating mtDNA without nuclear DNA;the method with SDS-proteinase K is suitable for isolat-ing the total DNA;the method with Trizol is suitable for preparing total RNA, total DNA and protein.
