猪线粒体DNA D-loop PCR-RFLP分析

Analysis of the Porcine mtDNA D-loop PCR RFLP

摘要: 为了应用RFL P分析猪线粒体DNA D-loop的多态性。应用PCR对西双版纳近交系小耳猪、广西巴马小型猪、贵州小型香猪、大约克夏猪、荣昌猪和长白猪血液总DNA样品中线粒体 DNA控制区(mt DNA D-loop)进行扩增,23种限制性内切酶消化,琼脂糖凝胶电泳检测。结果发现mt DNA D-loop扩增带清晰,与已知片段长度一致,RFL P分析未见长度多态。可见猪线粒体DNA D-loop酶切多态性贫乏。因此根据现用酶,应用mt DNA D-loop PCR RFL P分析,尚不能做猪品种品系鉴定。

Abstract: To analyze the polymorphism of the mtDNA D-loop in pigs,a pairs of primers were designed to amplify mtDNA D-loop of Xishuangbanna Small-ear pig, Guizhou miniature Xiang pig, Guangxi Bama miniature pig,Rongchang pig,Lang Yorkshire and Landrace. The amplification products were digested by 23 restriction enzymes and detected by 2% agarose gel. Clear amplification band were obtained,and the fragment length was the same as that of being reported before. No polymorphism was found among or in stocks. The results suggested that there is no polymorphism in porcine mtDNA D-loop RFLP,therefore it is not suitable as a method for the differentiation of the breed or stock in swine.
