Experimental Study on Climacteric in Rats

摘要: 以11~15月龄雌性SD大鼠为实验组,3~5月龄雌性SD大鼠为对照组,观察血清促性腺激素、性激素水平,阴道脱落细胞涂片,卵巢、子宫、肾上腺皮质光镜变化及心房肌特殊颗粒(ASG)和超微结构的变化。结果发现实验组大鼠阴道细胞涂片为无规则动情周期变化;血清雌二醇(E2)水平降低、促黄体生成激素(LH)水平升高,促卵泡生长激素(FSH)有升高趋势;卵巢组织切片显示原始卵泡及初、次级卵泡减少,退化卵泡增多,卵巢与子宫内膜变化不相匹配;肾上腺皮质总厚度及束状带变薄,球、网状带增厚,结构出现紊乱并有出血;ASG数目减少,心肌呈衰老趋势。结果提示11~15月龄普通级雌性SD大鼠已进入自然更年期,可用以研究更年期综合症。

Abstract: In experimental group of female SD rats at the age of 11-15 months and control group of those at 3-5 months, the serum levels of gonadotropin and sex hormones were detected, smear of vaginal cell, structure of ovary, uterus and adrenal cortex, changes of atrial specific granule (ASG) were observed by microscope and electron microscope. The results showed no regular estrous cycle in experimental rats, the levels of serum estradiol (E2) reduced and LH elevated significantly, FSH rose slightly, there is reduction in the number of the primordial, primary and secondary oogoniums with increase in degenerated oogoniums. The histological changes in the ovary and endometrium were not matched with each other,the myocardium showes the tendency of senile changes. The results suggest that the female SD ras at the age of 11-15 months enter their natural climacteric and perhaps it can be adopted as the animal model for human climacterie study.
