参芪扶心口服液对睡眠剥夺大鼠利钾尿肽和心功能的影响

Effects of Shenqifuxin Oral Liquid on Plasma Kaliuretic Peptide, Myocardial Contractility and Relaxation of Left Ventricle in Rats after Sleep Deprivation

摘要: 目的 探讨参芪扶心口服液对睡眠剥夺(Sleep Deprivation, SD)大鼠心功能的影响。方法 利用“小平台水环境法”(flower pot) 建立大鼠睡眠剥夺模型, 采用左心室插管术测定血液动力学指标; 放免法测定血浆利钾尿肽(KP)、心钠素(ANP)、血管紧张素Ⅱ(AngⅡ)及内皮素-1(ET-1)浓度。结果 与模型组相比, 高剂量组等容收缩末期心肌缩短最大速度(Vmax)、左心室舒张期压力下降最大变化速率(-dp/dtmax)及左心室内压峰值(LVSP)增加, 左心室舒张末压(LVEDP)降低, 血浆 KP、AngⅡ及ET-1浓度降低, 均有显著性差异(P<0.05或P<0.01)。中、低剂量组血浆ANP水平下降明显(P均<0.01)。结论 睡眠剥夺能够引起大鼠心功能障碍。参芪扶心口服液能降低睡眠剥夺引起心功能不全大鼠血浆KP、AngⅡ、ET-1浓度, 影响ANP分泌, 改善左室舒缩功能。

Abstract: Objective To observe the effects of Shenqifuxin oral liquid (SQFXOL, 参芪扶心口服液) on plasma kaliuretic peptide (KP) and the left ventricular remodeling and the myocardial contractility and relaxation of left ventricle in rats after sleep deprivation. Methods The hemodynamic parameters were measured by the method of the catheterization. Plasma concentrations of KP, ANP, AngⅡ and ET-1 were determined by radioimmunoassays. Results In high dose group, the LVSP, dp/dtmax and Vmax were increased; LVEDP was decreased, plasma concentrations of KP, AngⅡ and ET-1 were decreased significantly(P<0.05 or P<0.01). The levels of plasma ANP in middle dose group and low dose group were significantly lower than those in model group (all P<0.01). Conclusion SQFXOL can reduce the plasma concentrations of KP, AngⅡ, ET-1, and ANP, improve the myocardial contractility and relaxation of left ventricular in rats with HF after myocardial infarction.
