摘要: 目的 利用维甲酸(RA)和二甲基亚砜(DMSO)诱导猪胚胎生殖细胞(EG细胞)分化,证明猪胚胎生殖细胞能够在体外分化为多种组织细胞。方法 利用悬浮培养和无饲养层培养,将猪胚胎生殖细胞分化为拟胚体,再将拟胚体分别培养于添加维甲酸(Retinoic acid, RA)和二甲基亚砜(Dimethylsulphoxide, DMSO)诱导剂的培养基中培养,诱导胚胎干细胞分化。结果 当悬浮培养时猪EG细胞形成拟胚体,再经RA和DMSO诱导分化为类桑椹胚体、类ES样细胞集落、神经样细胞和滋养层样细胞。结论 猪胚胎生殖细胞能够在体外分化为多种组织细胞,证明猪EG细胞具有多能性。

Abstract: Objective Using RA and DMSO to induce porcine embryonic stem cells, they were certificated that could be differentiated into the various kinds cell. Methods Porcine embryonic stem cells were induced to embryonic bodies in culture of free feed or culture of suspension. Embryonic bodies were cultured in the medium with RA or DMSO respectively, to induce embryonic stem cell to differentiate. Results The embryonic bodies would differentiate into neuron-like cells, trophoblast-1 ike cells, morula-like colonies and ES-like colonies when they cultured in the medium with RA and DMSO. Conclusion Porcine EG cells could be differentiated into the various kinds cell. It was testified that porcine EG cells were pluripotency.
