人参皂甙Rg1慢性阻塞性肺疾病大鼠肺组织上皮间充质转化的影响

Ginsenoside Rg1 Alleviated Epithelial Mesenchymal Transition of Airway in Chronic Obstructive Pulmonary Disease Rat

摘要: 目的 通过烟熏法建立慢性阻塞性肺疾病(COPD)大鼠模型,用人参皂甙Rg1灌胃,研究人参皂甙Rg1对小气道上皮细胞上皮间充质转化(EMT)的影响。方法 烟熏诱导大鼠COPD模型,观察肺功能变化,肺组织HE染色检测大鼠肺气肿和小气道破坏情况。免疫组织化学、实时PCR检测肺组织E-cad和α-SMA的表达,观察人参皂甙Rg1对肺内EMT的影响。结果 人参皂甙Rg1可以减轻烟熏对大鼠肺结构破坏,改善肺功能,降低肺内EMT。结论 人参皂甙Rg1可能通过抑制EMT来改善COPD大鼠小气道纤维化。

Abstract: Objective To investigate the effect and the related mechanism of ginsenoside Rg1 in epithelial mesenchymal transition (EMT) in chronic obstructive pulmonary disease (COPD). Methods The COPD rat models were induced by exposure to cigarette smoke. Ginsenoside Rg1 was oral gavage dosed. The pulmonary function test and lung histologic HE staining were used to evaluate emphysema and small airway destruction. The expressions of E-cad and α-SMA were measured using immunohistochemistry assay, real-time PCR to evaluate the effect of ginsenoside Rg1 on EMT. Results Ginsenoside Rg1 ameliorated emphysema and airway destruction in COPD rats, improved pulmonary function, alleviated airway EMT. Conclusion Ginsenoside Rg1 can protest EMT in lung tissue,and may be act as potent intervention against small airway fibrosis in COPD.
