高糖高脂饲料联合地塞米松诱发树鼩血糖、血脂异常

Insulin-Resistance Tree Shrew Model Induced by High-glucose-fat-diet with Dexamethasone

摘要: 目的 用高糖高脂饲料+地塞米松(dexamethasone,DEX)诱导树鼩糖代谢和脂代谢紊乱,为建立实验性2型糖尿病疾病模型提供基础.方法 14只雄性中缅树鼩,随机分为高糖高脂组(胆6)和高糖高脂+DEX组(3 mg/kg,h.I.,n =8),连续观察8周,并测定树鼩空腹血糖(FBG)、体重、甘油三酯(TG),总胆固醇(TC),糖化血红蛋白(Halc).结果 高糖高脂饮食联合地塞米松皮下注射后能够在短时间内使树鼩血糖升高,体重降低,同时发生脂代谢紊乱,糖耐量异常.结论 高糖高脂+地塞米松连续多次空腹注射可诱发树鼩糖代谢、脂代谢紊乱,且动物状态良好.

Abstract: Objective To induce an impaired glucose and lipid metabolism model by high-glucose-fat feeding (HGF) plus DEX so as to facilitate type 2 diabetes study. Methods The model was induced by HGF and HGF plus DEX respectively in tree shrew. Fourteen normal male tree shrews were randomly divided into 2 groups: HGF group with 6 tree shrews and HGF plus DEX group with 8. Several param-eters in the tree shrews were measured. Results The fasting blood glucose,lipid level and OGTT significantly increased in HGF+DEX treated group compared with the HGF group while the fasting body weight reduced in both group. Conclusions High-glucose-fat feeding and DEX together can be used to induce glycometabolism and lipid metabolism abnormality.
