    Morris Water Maze Video Behavioral Analysis Using Imagej

    Objective To analyzed Morris water maze video based on the usage of ImageJ. Methods USB camera and common PC (Intel Pentium 4 CPU 1,70GHz) were used, and it's not necessary to mark the animals. After the procedures of setting scale, removing background and binaryzation with ImageJ, we used MultiTracker,an ImageJ plugin ,which can track the movement of multiple objects,to analyze the results. Result Swimming paths of rats and paths can be successfully analyzed. Conclusion This method based on a free software and low hardware requirements, can get all necessary date of Morris water maze.

    Objective To establish human lung cancer cell line and human liver cancer cell line with GFP/Luc double-tagging which having high expressing rate,and to observe development of the tumors in deep organisms of the nude mice with in vivo biofluorescence imaging. Methods The SPC-A-1-GFP/Luc cell line and SMMC-7721 -GFP/Luc cell line were established by lentivirus transfection, en-hancing their GFP/Luc double-tagging expressing rate to about 100% by flow cytometer, then were inoculated in lung and liver of the BALB/c nude mice respectively, finally, the behaviors of the two cell lines transfected stably in development of the tumours was detected by in vivo biofluorescence imaging.Results The cell lines of SPC-A-1-GFP/Luc and SMMC-7721-GFP/Luc with GFP/Luc double-taking have been built up with the expressing rate nearly 100%. After the two cell lines have been inoculated in lung and liver of the nude mice, the tiny tumor focuses in early time and the status of tumors in late time in the deep organisms were observed by the in vivo biofluorescence imaging. Conclusion The estab-lished cell lines with stable GFP/Luc double-tagging, coupled with the application of m vivo biofluorescence imaging, can detect the tumors in deep organisms of the nude mouse intravitaly,real-timely, quantitively,and may be offering an ideal device for researchers to further explore development of the tumor.

    Objective To study the growth and metastases of the tumors after orthotopic transplan-tation with hepatocellar carcinoma cell line (Huh-7) in nude rat. Methods Tissue from the tumor inoculated subcutaneously in nude mice with Huh-7 cell line was transferred to the nude rat beneath the skin. Subcutaneous and orthotopic implant models simulating hepatocellar carcinoma was established by suturing histologically intact human tumor tissue on the liver of nude rat. Subsequently,the rate of the orthotopic tumor and their biological properties of tumor growth and metastasis were observed.Tuinorgenecity, invasion,metastasis and morphological characteristics of the implanted tumors were studied by light microscopy, electron microscopy and flow cytometry. Results In orthotopic trans-plant model of nude rat,tumor formation rate was 90.0% and metastasis rate of liver was 50.0%.Conclusions The nude rat is more suitable for orthotopic transplant models for hepatocellar carcinoma,and should be valuable for research on mechanism and new therapies for metastasis.

    Objective To compare the biological character between the model of human prostate cancer cell DU 145 and 22Rvl utilizing ectopic and orthotopic implantation in nude mice. Methods The human prostate cancer cell DU 145 and 22Rvl were inoculated under orthotopic or ectopic environments separately in nude mice, the mice were were sacrificed at the agonal stage. The tumorigenicity and lymph node metastasis were determined by gross and histological examinations. Results The 22Rvl cells were equally tumorigenic with 10/10 frequency in the prostate and subcutis, they exhibited highly metastasis potentiality with 9/10 lymph node metastasis after orthotopic implantation in nude mice for 40 days, whereas no lymph node metastasis after ectopic implantation.The DU145 cells were equal tumorigenic with 10/10 frequency in the prostate and subcutis, they exhibited lowly metastasis potentiality with 1/10 lymph node metastasis after orthotopic implantation in nude mice, and no lymph node metastasis after ectopic implantation.Conclusion The lymph node metastasis model established by orthotopic implantation in nude mice is successful, and it may become a valuable model for the study of metastasis mechanism and experimental therapy of prostate cancer.

    Silkworm,i.e. Bombyx mori, as the typical representative of lepidopteran insect and the ideal biological model,has played a significant role in promoting the development of animal science, which specific mutation has already been the optimum material for the research on modern genetic engineering. With the accomplishment of the draft of the silkworm genome and the fine map, silkworm will further become a comparatively ideal model organism in the field of research on life sciences in postgenome era. This text summarized the silkworm’s contributions to the modem genetics; again, analyzed the status quo of silkworm as the model organism in researching realms of developmental biology, bioreactor, and the construction of molecular platform of genome and so forth; and finally, discussed the application of silkworm being the model organism in the field of Modem Biology.

    Objective To establish a human hepatocellular carcinoma cell line with stable and high-level GFP/Luciferase(Luc) expression, and observe the development of tumors in deep organisms of the nude mice by in vivo biofluorescence imaging. Methods The SMMC-7721 -GFP/Luc cell line were established by lentivirus transfection,then were orthotopic implanted in the liver of the BALB/c nude micc, the behaviors of the tumors was detected by in vivo bioluminescence imaging. Results The cell line SMMC-7721-GFP/Luc with stable and high-level GFP/Luc expression had been successfully established. After the cell line had been inoculated in the liver of the nude mice, the tiny tumor masses in early time and the growth of tumors in late time in the deep organisms were observed by in vivo biofluorescence imaging. Conclusion The use of cell line SMMC-7721 -GFP/Luc could directly observe the tumor growth and metastasis in the orthotopic implantation animal model. It would help us to understand the biological behavior of human hepatocellular carcinoma and its characteristics. In view of its sensibility, intuitiveness, dynamics and reliability, it would do some good to further study of human hepatocellular carcinoma.

    Objective To expand the application of toluidine blue staining. Methods Skin, lung,trachea, cerebrum and spinal cord tissues were collected and fixed by 4% formalin. After routine slicing,the slices were stained by toluidine blue. Lastly the feature of all kinds of tissues under microscope was observed. Results Besides nissl body was clearly showed,the langerhan's cell, melanocytes and merkel cell in skin can also be differentiated. The type I alveolarenhanced by toluidine blue staining.The layers of cerebral cortex were displayed much more clearly than HE stai cell,type II alveolar cell and macrophage in lung and the ciliated cell and goblet cell in trachea were markedly ning. Conclusion Toluidine blue staining may be explored in skin, lung, trachea, cerebrum and spinal cord tissues.

    Objective To construct a knockout HPRT targeting vector and a promoterless knockin targeting vector,which can be used for making rabbit HPRT gene knockout models and transgenic rabbit in the future. Methods The rabbit full length HPRT gene BAC clone LBNL1-304M19 is used as the template. A 12.5kb rabbit HPRT gene fragment,which does not include promoter, is cloned into pKS-plasmid to form pKS-rHPRT recombinant plasmid via Gap-Repair by Red recombination system. Then, the PCR was used to obtain the IRES-eGFPCre-Frt/Neo/Frt fragments flanking with 50bp homologous arms for homologous recombination of rabbit HPRT gene on the basis of pKS-rHPRT and pIGCN21 plasmids. A knockout HPRT taigeting vector by replacing the last three exons with a IRES-eGFPCre-Frt/ Neo/Frt cassette and a promoterless knockin targeting vector by inserting a IRES-eGFPCre-Frt/Neo/Frt cassette in the 3’UTR of the HPRT gene were rapidly constructed. Results The identification of the vectors with PCR,enzyme restriction and sequence showed two vectors were constructed successfully. Conclusions We rapidly constructed a knockout HPRT taigeting vector and a promoterless knockin targeting vector, the efficiency of deleting or inserting DNA fragment by homologous recombination technology has also been studied.

    Study on Biological Characteristics of Spontaneous Type 2 Diabetic db/db Mice

    Objective To compare the biological characteristics of spontaneous type 2 diabetic C57BL/KsJ-db/db mice in growth curve, glucose metabolism, serum insulin, weight of major organs,and organ coefficient with C57BL/Ks J-db/+ mice as the control, then the morphologic changes of the liver, kidney and pancreas were observed. Methods Forth weeks old mice were used with total of twenty weeks experimental period, the weight and glucose at each week, serum insulin at every two weeks were measured. After the experimental period, the mice were sacrified, and the organ and fat were weighted. The pathological changes of the liver, kidney and pancreas were observed. Results ① The weight, glucose and serum insulin of db/db mice were higher than those of the control group at the same age( P P<0.01). But the spleen,pancreas and kidney was less relatively, especially of the spleen and pancreas in female mice( P<0.01).③ The liver appeared significant cell putrescence, adipose degeneration, inflammatory cell infiltration and fibroplastic proliferation in db/db mice. The renal interstitial appeared hyperaemia, dropsy, hyaline cast in renal tubules, and adipose degeneration of epithelial cell. At the same time,the number of glomeruli pancreatici and islet cells decreased obviously. Conclusions The db/db mice is a suitable disease model of type 2 diabetes mellitus, which shows hyperglycemia, high insulin resistance,insufficiency of islet function, renal and hepatic alterations.

    Objective To establish a sleep apnea syndrome (SAS) model in rats. Methods Thirty two male Wistar rats were randomly divided into four groups: the control group, sleep deprivation group,intermittent hypoxia group, intermittent hypoxia and sleep deprivation group. The experiment lasted for 8 weeks, during the experiment, tail-cuff systolic blood pressure was measured every week. At the end of the experiment, invasive arterial blood pressure was measured and the cardiac and left ventricular were weighed. The Cardiac Mass Index was estimated. Tissues from the large artery, lungs and kidneys were embedded in paraffin and histological examination was conducted. Results During various intervals of intermittent hypoxia exposure or sleep deprivation, tail-cuff systolic blood pressure progressively increased.At end of the experiment, both the cardiac weight, left ventricular weight and invasive arterial blood pressure also increased, especially the intermittent hypoxia and sleep deprivation group. Compared with the control group, the arterial vascular wall, as well as the pulmonary arteriole vascular wall of rats in the intermittent hypoxia and sleep deprivation group became thicker. At the same time,epithelial cells of renal tubules showed typical hydropic degeneration and protein cast appeared in the renal tubules.Conclusion Asleep apnea syndrome rat model was successfully established by intermittent hypoxia exposure and sleep deprivation.

    Objective Histopathologic changes of lung from the pigs infected with highly pathogenic PRRSV was observed to explore the feasibility of pulmonary injury animal model induced by PRRSV.Methods Ten pigs of experimental group were infected with highly pathogenic NVDC-JXA1 PRRSV strain, and the lung tissues were collected for histopathologic observation. The pulmonary injury induced by highly pathogenic PRRSV and SARS was compared. Results Pigs of experimental group were all died after artificial infection of highly pathogenic PRRSV, and the typical changes of interstitial pneumonia can be found in lung tissue. There was a high similarity of pulmonary injuries induced by highly pathogenic PRRSV and SARS after comparison. Conclusion It was feasible that highly pathogenic PRRSV could be used as a model of viral pulmonary injury, which had a definite significance to study the pathogenesis of SARRS.

    Objective To explore a novel method for atopic dermatitis (AD) model in mouse induced by ovalbumin (OVA). Method The 6-weeks BALB/c female mice were subcutaneously injected with 0.5 mL of 200 µg/mL OVA solution on 1st day and subcutaneously injected with 0.5 mL of 100 µg/mL OVA solution on 6th day to induce systemic sensitization again.On 18th day and 39th day,1.5 cm 2 sterile gauze with 50 µL,1 000 µg/mL OVA solution were attached on the back of the mice and replaced every day for consecutive 7 days for the second local sensitization.On 46th day,the pathological changes in mouse skins were observed to judge whether mouse AD model was successfully established and the rates of being model and the mortality rate was calculated. Result The dermatitis changes,such as scab,flushing,desquamation,and so on,were shown in AD model mice.Also,the epidermal cells were thickened and the dermis was infiltrated with lymphocytes in AD model mice.The total serum IgE level in the AD model mice [(6 296.17±1 021.28) ng/Lsignificantly increased as compared with blank control mice [(1 280.97±501.24) ng/L]( P<0.01) and the mice injected with saline [(1 327.57±629.71) ng/L( P<0.01).The rates of AD model and the mortality were respectively 94% and 6%. Conclusion Our study introduces a simple and stable method for mouse AD model induced by OVA.

    Overdose of injectable barbiturates or barbituric acid derivatives is the most preferred euthanasia method for the majority of animal species. However, these controlled drugs have limited access, and in this case or in any other scientific concerns, veterinarians may choose alternative, conditional acceptable methods as recommended by American Veterinary Medical Association (AVMA) Guidelines for the Euthanasia of Animals. In the newest 2020 Edition, existing euthanasia methods, techniques and agents of euthanasia are updated according to recently released articles. This article summarized the updated content of the new version of the AVMA Guidelines and on this basis, enumerated the most preferred methods of euthanasia, methods acceptable with conditions and unacceptable methods to euthanize different animal species. In addition, principles and precautions related to animal euthanasia were also briefly described.

    Construction and Evaluation of Skin Photoaging Mouse Model

    Objective To establish a mouse model of skin photoaging induced by ultraviolet B (UVB) radiation, and to provide a reference for the study of skin photoaging. Methods Thirty male KM mice were divided into a control group and a model group. The control group received normal light, while the model group was subcutaneously injected with D-galactose combined with narrow-band UVB daily irradiation for 40 min at a dose of 120 mJ/cm2 for 40 d. The skin appearance of mice with fur on the back being removed was observed. Skin tissue pathology was observed after HE staining and Masson staining. Biochemical indexes of skin tissue homogenate including glutathione peroxidase (GSH-PX), superoxide dismutase (SOD), hydroxyproline (HYP), and malondialdehyde (MDA) were measured by enzymatic analysis. The expressions of matrix metalloproteinase-1 (MMP-1), c-fos, and c-jun were determined by Western blotting. Results The skin of the model group was darker, looser, drier, more rough, and deeper than that of the control group. Pathological observation revealed that the epidermis thickened and the dermis fibers were reduced, broken, and disorderly arranged in the model group. In the model group, the activities of GSH-PX and SOD decreased, the content of MDA increased, the content of HYP decreased, and the expression levels of MMP-1, c-fos and c-jun of the model group increased (all P<0.01). Conclusion The skin damage of mice induced by subcutaneous injection of D-galactose combined with UVB irradiation is consistent with photoaging, so it is an effective mouse model of skin photoaging.

    Objective To identify the role of new sperm-specific gene ssp411 in sperm maturation, fertilization, and embryo development, the expression vectors of ssp411 for RNA interference as siRNA-ssp411s were designed and contructed. Methods The interfering sequences of ssp411 were designed according to the sequence of ssp411 of GenBank.Three pairs of oligonucleotides were synthesized and inserted into plasmid pRNAT-U6.1 to generate siRNA expression vector, and were identified by PCR and sequence analysis. The recombinant plasmid siRNA-ssp411s and ssp411 expression plasmid ssp411-pDsRed was cotransfected into the cultured 293T cell line. The interfering efficiency was detected by real-time PCR. Results Three ssp411 siRNA frames were successfully inserted into the plasmid vector pRNAT-U6.1,and the recombinant plasmids were sent to the sequence analysis. The result of real-time PCR indicates that the siRNA expression vector which was designed by SSP411 protein coding area 3-22 got the best interfering efficiency as 71%. Conclusion The siRNA recombinant can be constructed successfully by RNAi technique for inhibiting ssp411 expression.

    Objective Establishing anger-out and anger-in inducing emotional diseases macaque model to provide suitable research object for brain center mechanism of anger-out and anger-in inducing emotional diseases. Methods Using female macaque and inducing anger emotion occurence, then applying “Emotional evaluation scale of female experiment macaque”to score different emotion type of macaque model. We will differentiate anger-out and anger-in types from all anger monkeys based on scale scores and finish evaluating different anger types. Using anger-out and anger-in monkeys individu-ally add to pressurization method to prepare anger-in or anger-out inducing emotional diseases monkey model. Results After modeling stress,anger factors of PMS liver-qi invasion model monkeys increased significantly than control team( P<0.05), while PMS liver-qi depression model monkeys had a notable change compared with control team( P<0.05). While there is no difference compared with control team ( P>0.05) after administration. Conclusion It could be inferred that target disease could be induced successfully using social surbodination stress according to results of 30-minute group ad libitum obser-vation and scale scores, subjective combined objective results were regarded as a good measure methods to evaluate target emotional disease occurence on macaque. Idea and method mentioned above provide a suitable research object for occurrence pathogenesy of anger-out and anger-in emotion inducing emotional disorder.

    Objcctivc The breakdance-mutation ( bretb 218) has been described as a mutation in the zebrafish orthologe of the human ether-a-go-go-related gene (HERG, kcnh2) with an arrhythmic heart beat in the homozygous. Interestingly, we found the heterozygous (hets) could generate multiple premature aging-like features and distinct electrophysiology. It made possible to develop a vertebrate model for study on premature aging associated to kcnh2. Methods Life span and body shape change were followed up and recorded. Exercise endurance capacity was examined by swim test. ECG test was performed in wild type (WT) and the hets at different adult stages. Results Survival rate dramatically descended and curly body showed from 20 months in the hets. ECG test showed heart rate dramatically decreased along aging process and QTc duration prolonged in the hets. Exercise endurance capacity significantly reduced 56.44% in the hets at 32 months comparing to WT at the same stage.Conclusion The heterozygous breib 218 has shorter life span and showed the presenility on appearance, electrophysiology and the exercise endurance capacity which may associate to the mutation in one allele of zebrafish orthologue of kcnh2.

The glucose and insulin of the fasting and 30min post of glucose injected of the all groups were determined, and the HOMA-β,△ INS30/△ GLU30 and HOMA-IR were calculated and observed. Results The glucose and insulin of the fasting and 30min post of glucose injected and the HOMA-IR of the S+LPS group were higher than that of the before scalded(P<0.01), but the HOMA-β and △ INS30/△ GLU30 of the S+LPS group were lower than that of the before scalded(P<0.01). The HOMA-β and △ INS30/△ GLU30 of the GIK group were higher than that of the control group, there was no difference of HOMA-TR significantly between the GIK and control group. Conclusion The serious burn rats represents dysfunction of early phase and basal insulin release of p cell, GIK treatment would be beneficial for the protect of β cell on this states.

    Objective To establish a mouse model of liver granuloma and investigate its mechanism. Methods C57BL/6J mice were injected Propionibactenium acnes (P.acnes) via tail vein. After injection, number of F4/80 -B220 -CD 11 c+ cells and level of ALT were detected at time points of 1,6 h,and 1,2,3,5 and 7 d. On the 7th day after injection, pathological changes of livers between treated mice and untreated normal mice were compared. Nad’ve mice were injected with the sorted F4/80 -B220 -CD1lc + cells from peripheral blood of P.acnes-treated mice to establish an adoptive transfer group. Results Number of F4/80 -B220 -CD11c + cells and level of ALT upgrade obviously after F4/80 -B220 -CD11c + cell inflision as well as P.acnes injection. Pathologic examination showed that granulomas were formed in the livers either in P.acnes or in F4/80 -B220 -CD1lc + cell induced mice, accompanied with inflammatory cells infiltration.Conclusion P.acnes can induce liver granuloma of mice successfully and F4/80 -B220 - CDllc + cell play an important role in liver granuloma development.

    Objective To explore the correlation between serum potassium levels and the changes of the heart, the real-time echocardiography was applied to monitor the effects of hypokalemia on the heart structure and function of the experimental rabbits, and for the purpose of early diagnosis and treatment in the clinical practice. Methods The hypokalemia experimental model was established by intramuscu-lar furosemide injection. M-mode echocardiography was used to monitor the cardiac morphology,the sequential variation of flow spectrum and cardiac function. After that, the heart was removed and the tissue was made into pathological section and dyed with HE. Results The cardiac morphology and function of the rabbits showed little change during the early and middle stage. Then, the heart dilated rapidly, and its function decreased until cardiac arrest. The myocardial pathological section showed swell and vacuolization of the myocardial cells. Conclusion There were a series of changes in the heart of rabbit during the decreasing process of serum potassium concentration, and echocardiography is a noninvasive method to monitor these changes, it can observe the heart morphology and evaluate the cardiac function accurately in real time. Thus, echocardiography provides a convenient and reliable method for monitoring the heart complications induced by hypokalemia.

    Objectctive To explore the protective effect of the recombinant MnSOD or CuZnSOD on the cryopreservation of wild mice sperm. Methods The R18S3 added with different concentrations of recombinant MnSOD or CuZnSOD was used as cryoprotectant to select two of the best concentra-tion of SOD through comparison of the motility,membrane integrity, DNA damage of frozen-thawed ICR mice sperm in each group. Results The motility and membrane integrity of frozen-thawed wild mice sperm increased significantly ( P<0.05) adding with 100U CuZnSOD and 200U MnSOD in R18S3 compared to the control group. While there were no difference in the group of R18S3 with 100 U CuZnSOD adding,only in the group of R18S3 adding with 200U MnSOD the fertility rate of frozen-thawed sperm significantly increased compared with the control group ( P<0.05). Conclusion The cryoprotectant adding with 200 U recombinant MnSOD has a protective effect on the cryopreservation of wild mice sperm.

    General anesthetics used for laboratory animals are mostly controlled drugs, and are subject to strict supervision by the competent government agency in China. Many general anesthetics recommended in the literature are either unavailable or difficult to procure/access in the market, resulting in limited options for clinical use. Furthermore, not all laboratory veterinarians have practical experience in species-specific anesthetic selection and use. Owing to these factors, general anesthesia presents a common institutional challenge in animal surgical programs and serves as a bottleneck that restricts the sustainable development of biomedical industries working with laboratory animal species. This article summarizes the pharmacological properties of common general anesthetics and provides suggestions for general anesthesia in different laboratory animal species.

    Objective To amplify and identify dendritic cells (DC) generated in vitro from porcine peripheral blood monocyte (PBMC) and study their immunological characteristics. Methods Monocyte-derived DC (MoDC) were generated by stimulating PBMC with granulocyte-macrophage colony-stumulating factor (GM-CSF) and interleukin-4 (IL-4),after 5 day culture,LPS (1 ug/ml) was added.Non-adherent cells were collected 5-8 days alter culture. Morphological features were characterized by phase contrast photomicrograph. The co-stimulation molecules, SLAII and other markers were assessed by FACS. Immunological functions were evaluated by means of mixed lymphocyte reaction (MLR) and ELISA. Results Porcine MoDC showed typical DC morphological features, phenotype were CDl +CD14 +CD80 +CD86 +CD172a +SLA Ⅱ +; the expression of CD80, CD86 and SLAII elevated after LPS stimulation. Porcine MoDC showed potent allogenic T cell stimulatory capacity inducing a strong interferon-γ and IL-4 response. Conclusion Porcine MoDC were successfully generated in vitro by stimulating PBMC with GM-CSF and IL-4, that might be meaningful for the further research on tolerance induction and xenotransplantation.
