摘要: RNA干扰作为一种有效的工具用来产生基因转录后沉默的效果,从而抑制特定基因的表达,已经得到成功应用.转基因RNAi小鼠的出现,使得该技术在哺乳动物中研究靶基因的敲低成为可能.本文综述了转基因RNAi小鼠的制备方法,转基因RNAi小鼠的应用并展望了转基因RNAi小鼠对功能基因组研究的贡献以及应用前景.

Abstract: RNA interference (RNAi)has been extensively used for sequence-specific silencing of target gene. The generation of RNAi transgenic mice made it possible to knock down gene expression in mammalian species. This paper reviewed the generation of transgenic RNAi mice and its application. The contribution of RNAi transgenic mice to functional genomics and of its prospect for application were also discussed.
