胶原酶冷消化法分离制备罗非鱼肝细胞

Preparation of Tilapia Hepatocytes by Cold Digestion of Collagenase

摘要: 目的 研究罗非鱼肝细胞的分离提取的方法、鉴定.方法 将鱼肝脏经D-Hank液反复洗涤后剪碎经IV型胶原酶消化后过滤、离心、经DMEM洗涤后用Percoll分离液行肝细胞纯化、细胞计数,光镜下观察其形态结构.结果 胶原冷消化法分离后的细胞中仍含有一定量的杂细胞,可采用90g,60g,30g离心进一步分离提纯.结论 胶原酶冷消化法获得满意结果,60g离心2min和30g离心3min可达到纯度和产量均较理想的结果.

Abstract: Objective To investigate the method of isolation and extraction of tilapia hepatocytes. Methods The fish liver was washing repeatedly by D-Hank, then the liver was cut into small pieces. The tissues was filtered and centrifuged after digested by Collagenase IV. Then the suspensions were purified by Percoll isolation medium after washed by DMEM. Hapatocytes was counted. The morphologic characteristics of the isolated hapatocytes were accessed and observed by light microscope. Result An amount mixed cells were found through the cold digestion of Collagenase method. Further isolation was done by centrifuged at 90 g, 60 g and 30g. Conclusion The method of Cold digestion by Collagenase had obtained the best isolated result. Better purification and amount of hepatocytes could be done by centrifugation with 60g for 2 minutes and centrifugation with 30g for 3 minutes.
