Objective
To observe the metastasis-associated genes expression in the highly metastatic human lung cancer cell line SPC-A-1sci and validate the highly metastatic characteristics of SPC-A-1sci
in vivo
.
Methods
The expression levels of several metastasis promoting genes concluding CD44, OPN, MMP-9, EGFR were analyzed between the high and low metastatic cell lines by RT-PCR, Western blotting, FICC, IHC and so on. The metastatic characteristics was investigated by injection the cell lines into the tail vein of nude mice.
Results
Compared with the low metastatic cell lines, the metastasis-associated genes CD44, OPN, MMP-9, EGFR in highly metastatic cells were significantly increased. After 8 weeks
in vivo
experiments, 100% (12/12) of mice injected with highly metastatic cell lines developed lung metastases, while low metastatic cell lines was 16.67%(2/12).
Conclusions
The metastasis-associated genes are highly expressed in SPC-A-1sci, and the SPC-A-1sci shows the typical characteristics of highly metastatic
in vivo
, and may be served as a reference for the application of the cell lines.

Objective
To evaluate application value of Sp3111 RNAi transgenic mice model and to study the function of the sp3111 protein in fertilization and early embryo development.
Methods
①real-time quantitative PCR was used to analysis the expression of sp3111 mRNA both in F1 and F4 generation, and comparing the suppression efficiency of RNAi between the two generations. ②Sperm samples collected from male Sp3111 RNAi transgenic mice(F4 generation) and from wild type mice as control group followed by IVF. The two groups were observed for the rates of fertilization and embryo fragmentation.
Results
①No significant difference was found between F1 and F4 generation in suppression efficiency of RNAi PŁľ0.05. ②The rates of normal and fragmented embryos of the experimental group were 34.48% and 46.21%, respectively,significantly different from those of the control group (
P
<0.05).
Conclusion
①the suppression efficiency of RNAi in Sp3111 RNAi transgenic mice could be transmitted stably to F4 generation. ②The low expression of sp3111 gene remarkably affected fertilization and early embryo development in mice.

Objective
To establish a myocardial ischemia model induced by ligation of coronary artery in WHBE rabbits,, and to investigate its cardiovascular system sensitivity through comparing to Japanese white rabbits.
Methods
Male WHBE rabbits and Japanese white rabbits were randomly divided into sham operation group and model group (n=10 in each). The sham operation group was only chest opened but not ligated and the myocardial ischemia model was induced by ligation of coronary artery. The degree of ST-segment elevation and activity changes of blood plasma AST, CK, LDH were observed before, at, and after making model for 60 min in each group, and myocardial infarction size was measured by NBT staining and image analysis software.
Results
Compared with sham operation group, the ST-segment was significantly elevated (
P<
0.01), and infarction size was also increased (
P<
0.01) , the blood plasma AST, CK, LDH activities were markedly raised after ligation of coronary artery in WHBE rabbits and Japanese white rabbits model group (
P<
0.05,
P<
0.01). Compared with Japanese white rabbit, the ST-segment, infarction size and blood plasma AST, CK, LDH activities in WHBE rabbits induced by ligation of coronary artery were slightly higher than those of Japanese white rabbits, especially for infarction size (

Conclusion
WHBE rabbit is more sensitive for myocardial ischemia induced by ligated coronary artery and ideal myocardial infarction model, and adapted to research of cardiovascular disease.

Objective
To rapidly detect the
Yersinia enterocolitica
in monkey ,a real time fluorescent PCR assay was established.
Method
The primers and probe were designed based on the conserved sequence of heat-labile enterotoxin gene of
Yersinia enterocolitica
. Then PCR reaction system was optimized and evaluated.
Results
The established real-time PCR method was sensitive and specific to 3.3×10 cfu/ml bacteria concentration. There was reliable correlation between the template copies (3.3×10

cfu/ml) and the Ct value.
Conclusion
The real-time PCR assay was proved to be specific with high sensitivity and efficiency, and would be a powerful diagnosis method of
Yersinia enterocolitica

Objective
To observe the expression of Bax and c-Fos in brain tissue of sub-health state rat model induced by restraint stress, and further to study the effect of the Traditional Chinese Medicine.
Methods
Twenty male SD rats of 320-360g, were randomly divided into 4 with 5 rats per group: normal control group, model control group, Anti-ageing tablets group and Liuwei Dihuang pills group, 5 rats per group. The sub-health state animal model was established by limiting daily movement for 3 hours consecutive to 14 days according to the circumstance that can result human physiological stress. At the same time, the rats in Anti-ageing tablets group and Liuwei Dihuang pills group were treated with Anti-ageing tablets and Liuwei Dihuang pills. Respectively during the model building period.The expression level of Bax and c-Fos in brain tissues were detected on completion of the model establishment .
Results
Fourteen days after molding, in model control group,the expression level of Bax and c-Fos in cerebral cortex and in hippocampus was increased remarkably, while expression of Bax and c-Fos in anti-aging tablets and Liuwei Dihuang pills treated groups was decreased significantly (
P
<0.05,

<0.01).
Conclusion
The rat with mental stress were induced by activity restriction have got the expression of Bax and c-Fos significantly increased in brain tissue . The treatments of anti-aging tablets and Liuweidihuang pills improve this kind of mental stress through reducing the expression of Bax and c-Fos, protecting and decreasing neuron injuries caused by stressors.

Objective
To observe the effect of melamine with different dosages on renal development in neonatal rats.
Methods
Fifty-six SD rats were randomly divided into 7 groups. groupM1-groupM6, fed with different dosages of melamine from the start of pregnancy, group M0 fed with distilled water at the same time. All neonatal rats on group M0 to M6 were sacrificed at the time of 12 h after deliver for conducting pathological examination on the kidneys.
Results
Compare to the control group (M0), the experimental groups showed different pathological changes with the different dosage of melamine.
Conclusion
By going through the placental barrier melamine could have a pathological effect on fetus kidney during pregnancy, and the degree of the effects relies on the dosage level of melamine.

Objective
To study the effect of mental stress on the alopecia areata incidence in C3H/HeJ mice.
Methods
Eighty C3H/HeJ mice were selected and randomly divided into 4 groups: mental stress group, drug-induced group, mental stress and drug-induced group, and control group, each group including 20 mice. Follow-up a period and observe the incidence of alopecia areata after intervention, and compare the differences among groups.
Results
The alopecia areata incidence of mental stress group was 10%, the control group did not occur; the incidence of mental stress and drug-induced group was 90%, incidence of drug-induced group was 60%, the difference was statistically significant (
P
<0.05).
Conclusion
The mental stress significantly increased the alopecia areata incidence of C3H/HeJ mouse.

Objective
To study the effects of water-solubility azone on the transdermal absorption of resveratrol and teratogenicity.
Methods
The formation of neovascularization was inhibited by using resveratrol, the number of zebrafish dorsal aorta intersegmental blood flow were calculated added with water- solubility azone under fluorescent microscope, and teratogenic-action effects were recorded by water-solubility azone added for 24 h.
Results
The water -solubility azone at the coneentration of 0.01~0.5 µg/ml were safe to zebrafish embry, and the 0.5 µg/ml water-solubility azone showed significant transdermal absorption of resveratrol in zebrafish, intersegmental vessel number of dorsal aorta inhibition ratio could be 22.9%, the incidence of deformity can be occured.
Conclusions
The water-soluble azone with the Concentrations over 0.5 µg/ml have significant teratogenic effects on zebrafish embryos; and the 0.5 µg/ml water-solubility azone shows the optimal transdermal absorption of the resveratrol.

Objective
To investigate the effect of a kind of Strong-bone Capsule on increasing bone mineral density (BMD) in castrated female rats, and explore its mechanisms.
Methods
Castrated female rats were treated with three different doses of the capsule (0.17, 0.33 and 1.00 g/kg) for three months, then the weight of rats, weight of the femur, the bone calcium content and BMD were examined; meanwhile, the calcium absorption experiment was also carried out on weanling rats with the same doses of each capsule, where the weight, body length, feces calcium content, ingestion content of the rats were measured and the uptake rate of calcium was calculated.
Results
The distal end and mid-point of the femur BMD in the moderate and high dose groups were significantly higher than that of the model control group; there were no significant differences of feces calcium content, ingestion content and uptake rate of calcium between the strong-bone capsule groups and calcium carbonate control group(
P
>0.05).
Conclusion
The capsule could increase the BMD of castrated rats by supplying calcium and regulating bone metabolism.

Objective
To observe the therapeutic effects of Shuangju Particles on experimental hyperplasia of mammary glands in rat and rabbit.
Method
The model of mammary gland hyperplasia was established by injection of estradiol benzoatics in this experiment, To observe the treatment of shuangju particles on mammary gland hyperplasia in rat and rabbit.
Result
The Shuangju particles can inhibit mammary enlargement caused by estrogen, reduce the level of E
2
in serum of rats and rabbit with mammary gland hyperplasia, increase the content of progesterone, reduce the number of lobular, acinar,the cell proliferative layer and the degree of mammary duct expansion. Meanwhile, the experiment of egg white induced foot swelling showed that it also has significant anti-inflammatory effect.
Conclusion
Shuangju Particles has preventive and therapeutic effects on experimental hyperplasia of mammary glands in rats and rabbit,and it may be used to treat disease of mammary gland hyperplasia in clinical.

The Goto-Kakizaki (GK) rat, one of the the best characterized animal models of spontaneous non-obese T2D, has similar progressive course with human. This animal is characterized by impaired glucose-induced insulin secretion, decreased beta cell mass, raised fasting blood glucose level, hepatic glucose overproduction and moderate insulin resistance in muscles and adipose tissues. Here, we review studies concerning genetic characterization, abnormal islet morphology, cytokines changes in some major tissues, such as islet, liver, muscles and adipose tissues, and the relationship of these changes and T2D etiopathology.

In this review, the molecular markers and genetic maps for zebrafish was introduced, the advantages and disadvantages of using these markers was discussed, and the polymorphism in zebrafish strains was described.
