Cryopreservation of Embryos From HBV Transgenic and Cataract Mice in a Vitrification Solution

摘要: 目的 研究开放拉长塑料细管法(OPS)对HBV转基因小鼠(P21HBxPHBx)和白内障小鼠(cataract mice,CM)胚胎冷冻效果.方法 采用乙二醇作为冷冻保护剂,以昆明小鼠为对照,用OPS法对CM及P21HBxPHBx两种小鼠3.5 d的胚胎进行玻璃化冷冻,并比较KM、CM及P21HBxPHBx获得胚胎数,解冻后胚胎的复苏率及发育率.结果 KM、CM及P21HBxPHBx小鼠平均获得胚胎数分别为31.12、19.32、9.14枚;解冻后的复苏率分别为95.6%、79.3%、80.3%;发育率分别为94.7%、73.8%、56.6%.表明HBV转基因小鼠的获得胚胎数明显低于昆明小鼠,解冻后三种小鼠的胚胎复苏率之间无显著差别,而解冻后发育率,P21HBxPHBx小鼠要明显低于KM小鼠.结论 转基因小鼠超排数及解冻后发育率低于昆明小鼠,而白内障模型小鼠和昆明小鼠比较不显著.

Abstract: Objective To study the value of open pulled straw(OPS) method on cryopreservation of embryos between HBV transgenic mice and cataract mice (CM). Methods Ethylene glygol (EG) is used as the cryoprotectant, OPS method is adopted to carry out vitrification freezing for 3.5d embryo of CM,P21HBxPHBx and KM mice, and compared the numbers of embryo,recovery rate and developmental rate after defrosting of KM, CM and P21HBxPHBx.Results The average embryo numbers of KM , CM,and P21HBxPHBx 31.12,19.32,9.14 respectively . The recovery rates are 88.7%, 81.1% and 80.3% respectively.The developmental rates are 82.6%,79.4% and 56.6% respectively. The embryo numbers ofP21HBxPHBx are lower than those of KM . The recovery rates are not significantly different in three kinds of mice .The embryo developmental rate after defrosting of P21HBxPHBx is lower than that of KM. Conclusion The superovulated numbers and developmental rate after defrosting 0f P21HBxPHBx are iower than those of KM,and there is no significant difference between CM and KM.
