高糖高脂高胆固醇致兔糖、脂代谢紊乱和胰、肝、肾组织结构的变化

摘要: 目的 探讨高糖高脂高胆固醇饲料对新西兰兔血糖、甘油三酯、总胆固醇水平和胰、肝、肾组织结构的影响。方法 将雄性新西兰兔20只随机分为2组,10只饲以基础饲料作为正常对照组(C组);10只饲以高糖高脂高胆固醇饲料作为实验组(HD组),共观察16周,每4周从禁食过夜的兔耳缘静脉抽取血样,测定血清中葡萄糖、甘油三酯和总胆固醇水平。16周时全部处死动物,取胰腺、肝脏和肾脏,用10%中性甲醛液固定,常规石蜡包埋切片,作HE染色、VG染色,并用免疫组织化学方法检测胰岛β细胞。结果 与对照组相比,实验组出现空腹血清总胆固醇、甘油三酯、葡萄糖相继升高(P<0.05)。以不同喂养时间作方差分析,整体上差异具有统计学意义。喂养16周后,实验组胰、肝、肾病理检查及胰腺免疫组化结果显示:胰岛数量减少,萎缩,胰岛内细胞数目亦减少,胞核浓染、缩小,胞浆内空泡变性明显,红染加深;肝脏脂肪样变性,纤维化;肾小球肥大,硬化,而正常对照组组织结构基本正常。结论 高糖高脂高胆固醇饲料对新西兰兔具有脂肪毒性和葡萄糖毒性,可致血糖、甘油三酯、总胆固醇水平升高,并可引起胰岛β细胞和肝脏、肾脏组织结构损伤。

Abstract: Objective To investigate the levels of blood glucose, triglyceride and total cholesterol, and pathological changes of pancreas,liver and kidney in New Zealand white rabbits fed with high sucrose, high fat and high cholesterol diet. Methods Male New Zealand White rabbits were divided into two groups: control group (n= 10,with normal fodder) and high sucrose, high fat and high cholesterol diet (HD) group (n=10, with 37% sucrose, 10% lard,2% cholesterol and 51% normal fodder), and observed for 16 weeks. Once 4 weeks after an overnight fasting, the blood samples of the rabbits were tested for the levels of serum glucose, triglyceride and total cholesterol. At the end of 16 weeks all the rabbits were sacrificed,and a part of pancreas, liver and kidney tissue were sampled for histopathological examination. Results The rabbits in the HD group emerged hyperglycemia, hypercholesterolemia and hypertriglyceridemia subsequently, and no obvious changes were observed in control group. The pancreatic islets from the HD group became atrophied, with loss in number of the islets and cells within them. The nucleus of islet cells were small and deep-dyed. There was an abundance of large vacuolations in cytoplasm. Hepatic steatosis and fibrosis, as well as glomerular hypertrophy and glomerulosclerosis, could be observed in the rabbits of HD group.It is suggested that high sugar, high fat and high cholesterol diet can increase the blood glucose, triglyceride and total cholesterol, and induce pathological changes of pancreas, liver and kidney in New Zealand White rabbits.
