    Analysis of Effects on In Vitro Fertilization and Embryo Transfer in Rhesus Monkey

    The percentage of normal morpha (68±10 to 82±7 and 84±6) and motility (72±11 to 85±6 and 83±9) of sperm could be improved by using swim up and density gradient centrifugation motile sperm separation methods, but there was no difference in fertility rate.136 oocytes were used in the experiment of IVF,30 of which were damaged to death during capacitation;the rest were inseminated and 56 of which fertilizated. 6 of fertilized oocytes were fixed, and 28 cleavaged. 11 of the 28 fertilizated oocytes developed to 8-cell embryos. Although the cryosurvival rate of three embryos was 100% after thawed, pregnancy rate was zero after being transferred. The results showed that Percoll and swim up motile sperm separation methods were effective, and both caffeine and dbcAMP are necessary for inducing sperm to capacitation, they have different effects on the ability of sperm to bind to the zona and to undergo the acrosome reaction. The optimal sperm concentration for IVF is 2×105 sperm/ml or so. Serum and hormone can improve the maturation rate of oocytes. And serum and amino acid are necessary for embryo development. The failure of pregnancy may be resulted from scarcity of recipient and other factors.

    To observe the effects of Raloxifene on bone mineral density, bone metabolism and lipid me-tabolism of ovariectomized rats. We detected the BMD of spine and total hip, serum estrogen, serum lipid, markers of bone turnover, Ca,P content in artery wall in OVX rats administered by Raloxifene.Result shows that the BMD of OVX rat treated with Raloxifene increased on the spine and total hip, the level of estrogen (E2) increased significantly, markers of bone formation (serum BGP) and resorption (urinary PYD) decreased significantly, in addition, total serum cholesterol as well as LDL-cholesterol decreased significantly, and serum HDL-cholesterol and triglycerides did not significantly changed. So the Raloxifene could protect metabolism of bone and lipid, possibly due to its estrogen-like effect.
    Effect of Calcium to Phosphorus Ratio in Its Utilization in Growing Laboratory Rhesus Monkey

    Forty healthy growing laboratory rhesus monkeys were randomly divided into A and B group. According to calcium to phosphorus ratio (1.2:1,1.4:1,1.6:1,1.8:1 and 2.0:1) each group was divided into five groups (A1,A2,A3,A4,A5 and B1,B2,B3,B4,B5 respectively),including 2 male and 2 female in each. The level of dietary calcium or phosphorus and calcium to phosphorus ratio were adjusted by CaCO3, CaHP03 and NaH2PO4. The result showed, that no effect of calcium to phosphorus ratio on its utilization in growing laboratory rhesus monkeys were observed in A groups (P>0.05). Calcium and phosphorus utilization of B1 group were the highest in B groups (P<0.01). The results suggested that the calcium to phosphorus ratio 1.2:1 with phosphorus level 0.89% in dietary was the best choice for calcium and phosphorus utilization in growing laboratory rhesus monkeys.

    To establish a mouse model of nonmyeloablative bone marrow transplantation, we used fe-male C57BL/6 (H-2b) mice as recipients and male BALB/c (H-2d) mice as donors.The re-cipients received Cyclophosphamide (200mg/kg) and total body irradiation (5 Gy).This regime is nonmyeloablative On the 36th day after transplantation,we detected the Y chromosome specific gene in bone marrow cells of recipients mice. We found that engraftment of allogeneic hematopoietic stem cells can be established under the nonmyeloablative pretreatment regime.We also found that the number of hematopoietic stem cells is very important for the success rate of making model.The rate of engraftment is 80% (8/10) when the recipients mice were given 2 × 107 bone marrow cells.

    To investigate the placental pathological changes after treatment of replenishing kidney formula,early abortion rat model was established by Bromocriptine at 6-8 gestation day,the model rats were treated with high and low dosage of the herbs and the progesterone as the positive control, the rats were sacrificed at 12 gestation day and the ultrastructure of the placenta were observed by electron microscope.The results find that compared with the normal control the basement membrane of thetrophoblastic epithelium of model group was thickened and tortuous, within the basement membrane there were lots of dense microspherical particles, mitochondrial showed coagulating and vacant, the cisternae of the rough endoplasmic reticulum(rER) were dilated and contained flocky material and also the degranulation of ribosomes on the rER were frequently found, the capillary endothelium was thicked with microvilli-like projections. In herbs and progesterone groups, smooth endoplasmic reticulum(sER) increased and dilated, there were plenty of mitochondria and some lipid droplet in cytoplasm, organell were normal mainly. The results suggested that the Bromocriptine could induce changes of placental structure and influenced placental normal function. Kidney replenishing herbs could reverse morphological alteration of ultrastrcture in abortion placenta,and converted to benefit in hormone synthesis, finally achieved the aim to prevent abortion.

    To investigate the therapeutic effect of 131I-iodinated oil internal radiation on glioma,131I-iodinated oil was injected sector-shapely into tumor model of C6 glioma in rat with arc-needle, mate hed with routine straight-needle injection,the therapeutic effect induced by 131I-iodinated oil intraleision radiation in the carcinoma models we rerecorded through survival time, body weight, the size of local carcinoma and detected by histopathological examination. The Results showed that the rats in arc-needle injection of 131I-iodinated oil in C6 glioma at subcutis could increase rat's survival time, the body weight kept less desent, the lumps necrosed wholely. Pathological and ultrastructural detection revealed that the cell necrosis and collapse, sever nuclear damage was observed in the death cells. The early characteristics of necrosis such as margination of heterochromatin was also found in some tumor cells. Besides, well differentiated tumor cells, deg enerative tumor cells and some lymphocytes were seen. Conclusion arc-needle inj ection of 131 I-iodinated oil step-by step and sector-shapely into tumor was a better method, necrosis was the major effect of 131I-iodi nated oil internal radiation in carcinoma at the level of treated dosage.

    The model of cerebral local infarcts in rabbits were established by means of photochemical induction. The results showed that the successful rate of the models were 98%, the focal infarct area was 0.465±0.012 cm2, the volume was 0.268±0.009 cm3,the infarct focus have showed typical pathological process of damage, exudation, inflammation. The rabbit models of cerebral local infarction established by photochemical induced methods were convenient, rapid and steady.

    In order to study the Rhubarb polysaccharide (RP) and its effect on some serum lipid in diabetic mice induced by alloxcan. Serum TG、TC、LDL、HDL、SOD and polyamine in pancreas tissue were detected.The results showed that the levels of TG,TC and LDL and also the polyamine content in pancreas tissue were decreased, whereas the concentration of HDL and activity of superoxide dismutatase increased in RP treated mice. The results suggested that the Rhubarb polysaccharide could improve the situation of serum lipid in diabetic mice induced by alloxcan.
