    The bipedal rats model was developed by forelimb amputation and special rearing methodvs.Normal rats served as the control. The ultrastructure of nucleus pulposus of L2-3 discs of these rats were observed under electron microscope. With the increasing of aging and stress, obvious degeneration and necrosis were observed in the notochordal cells in nucleus pulposus of SD rats. In the matrix the arrangement of the collagen fiber became disordered, lamellar structures and mass appeared. These results demonstrated that aging and stress accelerated the degeneration of nucleus pulposus of SD rats. The degeneration proce-dure was in agreement with that of human beings.

    To detect the liver spontaneous lesions of beagle dogs used in toxicity studies. Sixty beagle dogs from control experimental groups, age from 10-18 months were used. Liver fune-tion index of serum were tested and liver samples were observed by routine histological tech-nique. The results revealed that all 7 clinical index were deviated in different degree.Various spontaneous lesions amounting to 20 kinds in different degrees were found in 63.3% dogs of the study,including hepatocyte degenerations,necroses,and proliferations,connective fiber prolifer-ations and fibrosis,inflammatory cell infiltration,abscess and cystis. The degree and the frequen-cy of levsions was no difference between the sex. Usually several kinds of lesions occurred in the liver of one dog. It is suggested that spontaneous lesions should be monitored so as to provide histopathological investigations for safety estimation experiment of new medicine.

    The aim of this experiment is to develop a rat model with Thromboangitis Obliterans (TAO).The Wisar rats were treated with tobacoo sensitization,cold exposure, injection of TAO immune extract and altering of sexial hormone level, which are generally accepted as the important etiologic factors of TAO. Through observation of acromelic sign,pathological and electromi-croscopic scanning and hemorrheology examination,it was found all the changes occurred in these rat models were comparable to those of human TAO.
    L-NAME-Exacerbated Nephrosclerosis in Spontaneously Hypertensive Rats

    To evaluate whether N-nitro-L-arginine methylester (L-NAME ) can exacerbate nephrovsclerosis in spontaneously hypertensive rats (SHR),SHR were randomly divided into two groups according to their body weight. Group Ⅰ (SHR, n = 10) were given tap-water, while Group Ⅱ (SHR+NAME,n=9) were received L-NAME in the drinking water (50 mg/L).Twenty-four-hour urine samples were collected at baseline,weeks 2 and 4 after the start of L-NAME for proteinuria determination. Cardiac hemodynamic and renal clearance experiments were performed after the last 24 h urine collection. The severity of the renal injury was semi-quantitatively assessed. The results showed that the L-NAME in duced significant increases in mean arterial blood pressure and left ventricular end-diastolic prevssure at the end of the treatment ,but had little effects on heart rate and the maximal rate of rise of LV pressure (dP/dtmax). It also induced significant rises in the ratios of heart weight/body weight and left ventricle weight/body weight. In the kidney, L-NAME induced a progressive increases in proteinuria. It caused a profound elevation in renal vascular resistance, leading to a marked decrease in renal plasma flow, but a proportionately lesser decline in GFR. Thus, the filtration fraction rose. Se-mi-quantitative histological lesion grading revealed severe tubulointerstitial and renal vascular lesions,along with very mild glomerular sclerosis in SHR treated with L-NAME. Our results suggest that L-NAME exacerbated nephrosclerosis in SHR.Young SHR treated with L-NAME can he used as a model of chronic renal failure.
    Capacity of Immature Mice Oocytes to Undergo Maturation, Fertilization and Embryo Development in Vitro

    To Study in vitro maturation of GV stage oocytes from ovaries of pubertal mice,and compared their fertilization,embryo development with those in vivo matured,mechanical isolation GV stage and in vivo matured oocytes from pubertal mice. GV stage oocytes were cultured in the solution consisted of α-MEM+1mg/ml Fetuin + 10% FCS+1.5 IU/ml rHCG ±5 ng/ml rEGF,16h later were transferd to f-HTF+10% FCS for 4h for IVF,tlien cultured in M16+10% FCS for 72 h, recording maturation, fertilization and cleavage, and compared with oocytes in vivo matured. Results:The maturation rate of GV stage oocyte is 89. 68%. Further supplementation of rEGF was not significantly increased the maturation rate of GV oocytes (P>0.05),but the rate of fertilization, cleavage were significantly increased(P<0.05).The rate of fertilization and cleavage of in vivo matured oocyte was 62.83% and 60. 18%. However,the rate of fertilization and cleavage of in vitro matured oocyte was only 48.92% and 36. 69%,still lower than oocytes in vivo matured (P<0.01). Conclusions:GV stage oocyte of mice can be in vitro matured,fertil-ized,and cleavaged. Supplement the culture medium with Gn and rEGF promote the development potential of GV stage oocytes. However, the capacity of development of in vitro matured oocytes was lower than in vivo matured oocytes,indicating the culture system still need to be improved.

    Through water-absorbing function experiment and animal experimenr, comparing the effects of poplar shavings and general shavings to determine whether poplar shavings can be used as beddings. The water absorbed by poplar shavings is more than that of general shavings (P<0.05),the growth and breeding function between control group and experiment group is no different (P>0.05). It indicates that poplar shavings is safety,credibility,and should be expread ed for a large range which is used an the bedding for laboratory animal

    To determine the relationship between canine prostate volume and its weight,and to determine the value of transabdomen prostate volume determination with ultrasound,six beagle clogs and eight mongrel dogs were used. The volume of prostate were determined by ultrasound under anesthesia. After these dogs were killed by air embolism,their prostates were freed.The weight of prostates were measured and the volume was also measured by draining water method. Results showed that the volume detected by ultrasound approached to the practical prostate volume (r1 = 0. 920, r2= 0. 992 respectively), and the prostate vol-ume and weight had close relation-ship (r3 = 0. 990, r4= 0. 992). Therefore, it is very convenient to know canine prostate volume through ultrasound method,and according to the relatively constant prostate density,the prostate weight and volume can be converted each other.

    To investigate the regulatory role of osteogenic growth peptide (OOP) on expression of Type I collagen, osteocalcin and alkalin phosphatase mRNAs and ultrastructural features in rat osteoblast-like cell. Cultured osteoblast-like cells were divided into experimental group treated with OGP and control group. Expression of Type I collagen mRNA,osteocalcin mRNA and alkaline phosphatase mRNA of cells were determined by RT-PCR method. Expression of mRNA of cells can be got by using glyceraldehyde-3-phosphate dehydrogenase (GAPDH) as the value of in-ternal reference. Ultrastructural changes of cells were observed under transmission electron mi-crescope. The Rough endoplasmic reticulum of cells were more abundent in the OGP group than that of in the control group. Swelling end of tubby rough endoplasmic reticulum turned to pool and ribosome on rough endoplasmic reticulum was observed in the OGP group. Differentiation of cells was excellent in the OGP group. Expression of mRANs of cells were higher in the OGP group and relatively lower in the control group. Expression of Type I collagen mRAN and osteocalcin mRNA of the cells in the OGP grouop were gradually upregulated with increasing concen-Irations of OGP from 10-12-10-8 mol/L. The results revealed that the OGP can promote differentiation of cells and the Expression of Type I collagen mRAN,osteocalcin mRNA and alkaline phosphatase mRNA of the cells can be upregulated by exogenously administered OGP. Therefore ,OGP can promote the function of bone formation in rat.

    To study the law of body weight change of Glucocorticoid rats ,The Grey model GM (1, 1) was used to analyze and forecast the change of the rats body weight. The foreccust precision of GM (1,1) was high for the bodyweight change and the results were reliable,so the GM(1,1) could be used to forecast the bodyweight change of Glucocorticoid rats.

    To establish the rat model of Chemotherapy-induced anorexia (CIA) with cisplatin (DDP),the food consumption,the body weight and stool had been observed for 4 days. Then the rats were injected ip DDP(0. 5mg/ml)5mg ·kg-1 at the fifth day, thereafter the rats were in-jected DDP once again.The body weight,the gastric mucosal lesion score, gastrointestinal tract hematoxylin-eosion staining and 5-hydroxytryptamine (5-HT) expression were also determined. The rat’s food consumption was decreasing immediately from the first day of injection DDP, and some of rats even ate nothing. But at the fourth day the food consumption began to increase,we found there were gastric mucosal lesion, weight loss, inflammation in gastrointestinal tract,rise of 5-HT expression in model group. It was suggested that the rat model of CIA can be established by using DDP.

    The weight of main organ of 60 rhesus monkeys were measured and their coefficient were calculated.In the same age, there are no sex difference of organ weight and coefficient in juvenile group,and the weight of heart,spleen,brain, pancreas are heavier in male than female in both adult and senior groups,but the organ coefficient is no sex difference in those two groups. All the organs except for the thymus, the weight are increased with age, especially the heart,liver,spleen, lung, kidney, testis and prostate.Most of the organ coefficient are no age related.
