    Histopathological Observation on Spontaneous Lesion of Digestive System in Macaca mulatta

    To study the histopathological changes in digestive system spontaneous lesions of breeding macaca mulatta,the tongue, cheek pouch, esophagus, stomach, intestine, salivary glands, gall bladder and pancreas samples from the monkey of 2~20 years old were investigated by routine histological technique. The results showed that various spontaneous lesions including inflammatory cell infiltration, degenerations and necroses in mucosa,atrophy and proliferations in digestive glands, totaling 32 kinds in different degrees, were found in 92. 3% of the monkeys. The organ total detectable rate is higher in large intestine,check pouch and small intestine,and lower in esophagus and salivary gland. The degree and the frequency of lesions increased with age. It is suggested that spontaneous lesions should be monitored in safety estimation experiment of new medicine.

    To investigate the roles of pancreatic and peripancreatic tissues in acute pancreatitis,the rat model of acute biliary pancreatitis was established by retrograde injection of sodium taurocholate into the pancreatocholangitic duct and the primary and secondary lesions as well as pancreatic pathologic findings were observed. 1. Gross pathologic findings: at the same interval after model establishment, differences between the groups were very significant (P<0.01). A positive correlation existed between the severity of pancreatic and peripancreatio pathological changes (P<0.01). 2. Microscopic pathologic findings:at 6 hours,the necrosis of interstitial hemorrhage and fat differences between the groups was significant (P<0.05),and the other lesions were significantly different (P<0.01). 3. Exceptional pathologic findings:in two cases, mild pancreatic edema or necrosis with extensive infiltration in the retroperitoneal space. In three cases, severe pancreatitc edema or hemorrhagic necrosis with localized retroperitoneal infiltration. In five cases, hemorrhagic necrotizing model with severe lesions of splenic hilum. Under comparable conditions of model preparation, the severity of primary lesions is parallel to that of the second ary lesions. The discrepancy of the local lesions may be related to the differences of local tissue structures.

    To provide a series studies for influence of the left lung inverse circulation and BCPA on pulmonary physiology and function and probably to use this technique to treat the patients with functional single ventricle with pulmonary hypertension, the reversal model of left pulmonary circulation (RLPC) with bidirectional cavopulmonary anastomosis (BCPA) was established in canines. Six mongrel dogs were chosen to be studied on this model. The procedure of establishing RLPC plus BCPA include anastomose the distal end of left pulmonary artery (LPA) to the left auricle; use a graft tube to connect the proximal LPA to three pulmonary veins and do a end to side anastomosis between supra vena cava and right pulmonary artery.All canines were survived over three days. Echo and angiography showed that there were no remarkable obstruction in left lung inverse circulation and cavopulmonary anastomosis. So the RLPC plus BCPA is technically feasible,it affords a experimental model for in vivo study of pulmonary pathophysiology and haemodynamics in complex congenital heart diseases kids with pulmonary hypertention and it’s a interm palliation and can be transit to total cavapulmonary shunts.
    Establishment of the Model for Acute Vascular Rejection in Heart Xenotransplantation

    To establish the model of acute vascular rejection in heart xenotransplantation,the Guinea pigs and SD rats were served as donors and recipients respectively. Heart from the donor was transplanted heterotopically into the recipient,the aorta of the donor heart was anastomosed end-to-side to the abdominal aorta of the recipient and the pulmonary artery was anastomosed end-to-side to the inferior vena cava. To prevent hyperacute rejection,Cobra venom factor (CVF) was given intravenously at a dosage of 60U·kg-1 preoperatively,same dosage was given by an i. p. injection per 8 hours postoperatively. To prevent cellular rejection, FK506 was given at a dosage of 0.25mg·kg-1·6h-1 by an i. p. injection per 6 hours postoperatively. 18 operations of transplantation were done with the successful rate of 88.9%,the mean survival time of the xenografts was 24±3.05 hours. The histopathology of the rejected xenografts showed diffuse in-travascular thrombosis, interstitial hemorrhage associated with a large number of infiltrated inflammatory cells, focal ischemic infarcts and coagulative necrosis,consisted with the histological description of the acute vascular rejection reported by others. We could draw the conclusion that the guinea pig-to-rat heart transplantation model could be performed without microscope,its simplicity, easiness and reliability suggest that it is a good model in studying acute vascular rejection in heart xenotransplantation.

    Experimental ascending acute pyelonephritis was produced in rats by injecting 0.5 ml of 106/ml or 108/ml Escherichia coli O111B4 into the urinary bladder via. without unilateral ligation of the ureter. Results of the research showed the pathologic changes of kidneys were not so serious and no purulent were found,but the process of inflammation was typical.Two groups of the Escherichia coli O111B4 all could produce acute pyelonephritis and did not show any side preference.

    To examine the pathogenesis of pregnancy-induced hypertension syndrome(PIH) in rat and the feasibility of treating it with L-arginine. The rat PIH model induced by methyl ester was established, and treated with L-arginine. The therapeutic efficacy on rat PIH and the changes of the indexes such as NO, NOS, ET, ANG II,TXA2 and PGI2 were detected. The symptoms and physical signs of rat PIH were obviously ameliorated by administration of L-arginine. Compared with control group,low dosage group and high dosage group,the content of serum NO,ANG II and PGI2,the activity of rat NOS in model group were decreased, while ET,and TXA2 increased.So,the pathogenesis of PIH are the decrease of level of NO, activity of NOS and dise-quilibria of blood vessel regulating factors. The fact that the L-arginine is effective for PIH may provides a new reasonable and feasible way for PIH therapy.

    To study the celluar immune responses of HBV transgenic (Tg) mice induced by genetic recombinant Hepatitis B vaccine,the HBV transgenic mice were immunized by Hepatitis B vacc-ine,and killed at different period. The spleen were removed, and lymphocyte were separated and divided into two parts. One were cultured for 48h with HBsAg and assayed for IL-2 and IFN-Yand the other part were labelled with 3H-TdR after cultured for 56 h,and cultured for another 16h. The culture were harvested and the cpm was determined by using a liquid scintillation counter. The result showed that the Thl cytokine production and lymphocyte proliferation of HBV Tg mice after repeated immunization for 8 months were much higher than those of immunized one time for 1 week. But after immunized as normal style with different doses,the IL-2 level were not different to one time immunization. The results suggested that the high dose with repeated immunization might broken the immunized tolerance of HBV Tg mice and upregulate cella-ar responses.

    DNA extracted from Brucella (Rough、Smooth) and five non-Brucella micro-organisms were amplified by PCR using 2 pairs of primers specific for the genes encoding a 31-kDa Brucclla protein. Two DNA fragment of the expected size (591 bp and 460 bp) were amplified from Brucella (R、S) and one DNA fragment of the expected size (460 bp ) was amplified from Brucella using NT-PCR ,but no band was observed in five non-Brucella micro-organisms. The sensitivhy of the reaction was determined with different concentrations of genomic Brucella (R、S) DNA. As few as 1 pg DNA could be detected by this method.

    To establish a stable and repeatable heterotopic rat heart transplantation model,the classical Ono’s procedure was modified and the factors which influence the success of transplantation were explored in this article. The modification is that the donors’ brachiocephalic trunk and right pulmonary artery were anastomosed end to side with the recipients’ abdominal aorta and inferior vena cava respectively. In contrast to the classical operation, the occlusion time of the inferior vena cava and abdominal aorta was significantly shortened,and the time of cold ischemia was also shortened in the improved technique. Furthermore,the amount of bleeding also decreased. The improved technique showed superior to the classical Ono’s procedure and is recommended to be used more widely.

    To investigate the weight-reducing effect of methyl cellulose M450 on aged ovariectomized (OVX ) rats,the cellulose was administered intragastrically to ten-month old rats that were ovariectomized in six-month for 30 days, the body weight, weight of celiac fat tissue,adipocytc number,adipocyte size,blood lipid and cholesterol were observed. The results showed that the body weight,weight of celiac fat tissue,adipocyte size in the rats of 100 mg/kg and 200 mg/kg cellulose groups were significantly lower than those in control group (P<0. 05); and the level of blood lipid and cholesterol were lower too in the 200 mg/kg group (P<0.05);the results suggested that administration of cellulose (>100 mg/kg) to rats for 30 days have evident weight-reducing effect and can also decrease the blood lipid level, but 30 mg/kg cellulose group have no effect at all (P>0.05).

    The weight of main organ and their coefficient of 62 beagle dogs were measured.The results showed that in young age group,the weights of heart,lung and kidney were higher in male than in female. Both male and female, the weights of lung, thymus and thyroid in middle age group were higher than those of young age. The coefficient of the lung, thymus and thyroid were significant difference between the young and the middle age group,especially the difference of thymus coefficient was very significant.

    To explore the genotoxicity effects of passive smoking on Wistar rats,micronuclear test in polychromatic erythrocytes (PCEs) of bone marrow and sperm morphological test on Wistar rats were performed. The sperm malformation rate and the micronuclear rate in passive smoking groups were significantly higher than those in negative controls(P<0.05). The results suggested that the passive smoking not only affects the body cell but also reproductive cells of Wistar rats.

By reviewing related literatures ,the authors think that the an-imal models need further consummation in order to be applied in experimental study better. This is the fundamental assurance for assessing the action of the prescription and its mechanisms accu-rately.
