    Study on the Rescreening of 10 Genes' Functions Using Different Laboratory Mouse Strains

    Employing the technology of antisense oligo-nucleotide acid,the authors established a mouse behavior monitoring platform aimed at the large scale screening of novel gene functions in CNS. And with this platform, 38 genes have been screened using BALB/c mice. Among them, 10 genes showed differences to the control groups (saline,scramble). The authors screened the 10 genes again with C57BL/6 and ICR mice, and found the appearances were not always the same in three strains. In the first round screening, there are 6 of the 10 genes related to memory, 2 related to pain sensitivity,and the other 2 related to locomotion activity. But after the rescreening, only 3 genes show relativity with memory (Am , L, and Sn),and 2 genes show relativity with pain sensitivity (Pe, Rp). The results indicate that the importance of using different strains in the experiments, and it is better to use inbred strains than outbred colonies in the screening of gene functions. The results suggested that the research on different appearances of different mouse strains in one experiment and the use of different mouse strains to testify one result are both having very important significance to animal experiments and laboratory animal science.

    Ca2+currents were obtained in acutely dissociated mouse spermatogenic cells using whole-cell patch clamp technique. The results indicated that inward currents were recorded when K+ currents were blocked by Cs+and TEA+stepping membrane potential to voltage between —80 and +10 mV,in 10 mV increments from a holding potential of —90 mV,the peak amplitude occurred at between —30 mV and —40 mV,and the inward currents remained unchanged in the presence of 4 μmol/L tetrodoxin (TTX),a special sodium channels blocker,suggesting that it was generated by opening Ca2+channels other than Na+channels.

    To screen out the steady and controllable animal model and study the effect of the compound TCM on the degree of intestinal adhesion in rats after surgery,the SD rats of intestinal adhesion model alter surgery fed with the compound TCM Huanghou Paiqi Granule at the dosage of 2.5、5.0、10.0g·kg-1·d-1 continuously for 7 days for studying its effect on the degree of intestinal adhesion. The Results showed that the caecum model was easy to establish,steady and controllable, the Compound TCM could obviously improve the degree of intestinal adhesion in rats after surgery.

    To examine whether rat osteoportic model was established quickly by high protein and low calcium diets, Forty female Wistar rats (3 weeks old)were randomly divided into 5 Groups and fed with diets formulated according to AO AC for twenty weeks. The contents of protein and calcium in 5 Groups were 18%,1%;8%, 0.025%;8%;1%; 27%,0.025%;and27%,1% respectively. The results showed that the content of bone calcium in Group 2 and Group 4 was obviously decreased compared with that of Group 1 (P<0. 001), the BMD and hending stress resistance of femur was also decreased (P<0.01), and the trabecular area of lumbar and tibia was increased. In compared with Group 1, The body weight gain of Group 2 was obviously decreased (P <0. 001) and estradion in serum was decreased (P<0.05). The results suggested that the Rat osteoportic model established by high protein and low calcium diets for twenty weeks was successful.

    To study the changes of Mysion Heavy Chain (MHC) and myocardial structure in heart failure rats induced by doxorbincin (ADR) , 22 male SD rats were divided into ①control group (n= 11) and ②ADR group (n= 11, ip 20 mg/kg ADR given intermittently within 16 days in 8 times). Body weight of the rats,left ventricle mass to body weight ratio(LVM/BW),α-MHC and β-MHC were measured. The ultrastructural changes in myocardial cell were observed electromicrographically.Shifts of myosin from α-MHC to β-MHC occurred in ADR group in a form of the decrease of a-MHC (vs control P<0.01)and the increase of β-MHC(vs control P<0.01). In ADR group, an increase in LVM/BW is found (vs control P<0.01). Ultrastructural changes of myocardial cell in heart failure rats were electromicroscopically observed as lysis of myocardial filament extension and aelveoli of plasmalemma, crests rupture and swellening of mitochondria in ADR group. The results suggest that the MHC may be involved in myocardium remodeling.

    To elucidate the importance of intact animal model in pharmacological study of antiinflammatory-immunomodulating drugs (AIIDs) , some progress of intact animal models in antiinflammatory-immunomodulating pharmacology were reviewed,such as animal model of adjuvant arthritis, systemic lupus erythematosus, immunological liver injury,acquired immunodeficiency syndrome, Alzheimer disease,transplantation immune, immuno-neurologic disfunction and mucosal tolerance, and consequently to provide new ideas on pharmacological study of AIIDs and therapy of inflammatory and immunological diseases.

    Between 1970 and 2001,eleven coat color mutants of Mongolian Gerbil arose in scientific laboratory and pet population. Among them, eight coat color and their genetic mechanism were detailed in this article. The wild system includes two loci, one is A locus (coat color is Agouti) , another is a locus (coat color is Non-agouti or black). C system includes c locus (Albino) , ch (Acromelanistic albino),cchm (chinchilla medium). The white spotting mutant is controlled by Sp locus, g locus appears to a gray coat color, and p locus seems to be master of pink-eye dilution.The hairless phenotype is only elementary introduction.
