两种方法建立2型糖尿病肾病大鼠模型肾损伤相关指标的对比分析

Comparative Analysis on Renal Injury Relevant Indicators From Two Type 2 Diabetic Nephropathy Rats Models

摘要: 目的 对两种2型糖尿病肾病大鼠模型的建立方法及肾损伤相关检测指标进行对比研究和分析。 方法 利用三联法(切除左侧肾脏+高糖高脂饲料喂养+腹腔注射链脲佐菌素)和二联法(高糖高脂饲料喂养+腹腔注射链脲佐菌素)建立两种2型糖尿病肾病大鼠模型,并检测大鼠模型空腹血糖、胰岛素敏感指数、n-乙酰-β-d-氨基葡萄糖苷酶、非酶糖基化、肌酐、尿素氮、尿微量白蛋白、肾重比,病理学观察大鼠肾脏组织学变化。 结果 ①与正常对照组大鼠对比,三联法组和二联法组大鼠空腹血糖、n-乙酰-β-d-氨基葡萄糖苷酶、非酶糖基化、肌酐、尿素氮、尿微量白蛋白、肾重比明显增加,而胰岛素敏感指数明显降低,肾组织学显示明显的损伤改变,其中三联法组大鼠死亡一只.②与三联法组大鼠对比,二联法组大鼠空腹血糖、胰岛素敏感指数、n-乙酰-β-d-氨基葡萄糖苷酶、非酶糖基化、肌酐、尿素氮、肾重比没有明显差异。 结论 三联法和二联法均可以建立2型糖尿病肾病模型,这两种方法结果之间没有明显差异。从模型成功率、成活率,以及实验操作方便性方面考虑,二联法优于三联法。

Abstract: Objective To compare two methods to establish type 2 diabetic nephropathy rats models and the renal injury indices. Methods Type two diabetic nephropathy rat model was established with three-unite-method (STZ,fat-diet, unilateral nephrectomy) and two-unite-method(STZ, fat-diet). Fast-ing blood glucose, insulin sensitivity index,NAG,AGEs,SCr,BUN,mAlb, kidney index were measured respectively and renal pathological changes were observed. Result ① Compared with normal control group, rats in three-unite-method group and two-unite-method group present hyperglycemia,insulin resistant, some characteristics of diabetic nephropathy and renal damnification was observed.② Compared with the fasting blood glucose,insulin sensitivity index,renal function of three-unite-method group,those in two-unite-method group were unconspicuously different. Conclusion Both three-unite-method and two-unite-method were the suitable methods to establish type 2 diabetic nephropathy rats model.
