摘要: 目的 以重组禽干扰素α(IFNα)为基础,针对禽病毒性疾病的预防和治疗,研制禽用抗病毒制剂。方法 将禽干扰素α、胸腺肽α等进行基因重组改造,结合先进的发酵工艺,在E.coli和Picha pastoris高效表达系统中分别表达,再分离、纯化。结果 不同载体和不同菌株表达的IFN-THY融合蛋白的IFN活性测定结果有一定的差异.96 hIFN活力在0.13×104~8.12×104IU/ml,110h为1.22×105~3.27×106IU/ml。结论 48~110h不同样品均有明显的THY活性。

Abstract: Objective Based on recombination IFNA6 (Chicken interferon alpha), anti-viral biologic products were developed, aiming at prevention and treatment of poultry viral disease. Method Chicken interferon alpha(IFN) and thymosin alpha(THY) were recombined and expressed in E.coli and Picha pas tor is. Results There were different IFN biological activity from fusion protein of Chicken interferon and thymosin (IFN-THY) expressing in different vectors and different strains, 0.13 X 104-8.12 X 104IU/ml at 96 hours, and 1.22 X 105-3.27 X 106 IU/ml at 110 hours. Conclusion There were significant THY biological activity from different samples at 48-110 hours.
