Study on the Acute Brain Aging Rat Model by Radiation

摘要: 目的 根据衰老的自由基理论,建立一种简便易行的脑老化模型。方法 以60Co照射大鼠建立急性脑老化模型、D-半乳糖注射大鼠建立亚急性脑老化模型,5月龄大鼠作为青年对照组;通过Y型迷宫测定大鼠的学习与记忆能力;外周血SOD和MDA检测大鼠的抗氧化能力及脂质过氧化情况;通过光镜观察脑组织神经细胞的丢失、坏死及退行性变。结果 Y型迷宫测定表明,两个模型组大鼠的学习和记忆能力比对照组明显下降;与对照组相比,外周血中SOD明显降低,而MDA则明显升高;光镜下,在模型组不同脑区均能观察到神经元的丢失和坏死,以海马和皮质最明显。结论 成功建立了一种简便易行的急性脑老化模型;为脑老化机制的研究提供了新的实验手段。

Abstract: Objective According to the free radical theory for aging, to establish the simple and easy method for brain-aging model, providing theory for brain-aging mechanism and clinic prevention and therapy. Methods The acute brain-aging model was established by 60Co radiation, the second-acute one was established by injection of D-gal, and 5-monthly SD rats as the young control group. The rats, learning and memory ability was determined by Y type labyrinth, the rats’ antioxidant and lipid peroxidative injury were measured by SOD and MDA in blood. The neuron’s lose, injury and degeneration were observed with microscope. Results We found the descend of brain aging rat’s learning and memory ability with Y type labyrinth;The blood’s SOD in brain-aging model was significantly lower than that of the control, the MDA content in brain-aging model was markedly higher than that of the control; The neuron’s lose, injury and degeneration were observed by microscope in different brain region of the rat model, especially in cortex and hippocampus mostly. Conclusions The acute brain-aging rat model can be established simply and easily by radiation.
