Detection of Rodent Coronavirus by Using Reverse Transcriptase Polymerase Chain Reaction

摘要: 为了检测啮齿类动物冠状病毒,设计了特异性引物,建立RT-PCR检测方法,并用于检测。在基因文库中查找了啮齿类动物冠状病毒和SARS病毒的基因序列,从N基因片段中,利用计算机软件设计了一对特异性引物,通过确定反应混合物的浓度和反应条件,并建立了一整套从病毒RNA抽提、反转录到PCR反应的快速检测啮齿类动物冠状病毒的方法,扩增出了148bp的目的片段。并已用于啮齿类动物中检测各组织和拭子中的冠状病毒。该方法对啮齿类动物冠状病毒的早期诊断和与SARS病毒区别有重要意义。

Abstract: In order to detect rodent coronavirus, a pair of PCR primers were designed according to the compared sequences of rodent coronavirus and SARS coronavirus. The primers derived from the specific and conservative N gene,RT-PCR method was established. Result of rodent RT-PCR amplification showed the amplified products of 148bp length; RT-PCR method of rodent coronavirus has been used to detect the samples. The rodent RT-PCR method is important to determine early rodent coronavirus and discriminate SARS coronavirus.
