大鼠海马神经元急性分离和膜片钳方法的研究

Study On the Method of Acutely Isolating Hippocampal Neuron in Rat and Patch Clamp

摘要: 应用酶消化及机械分离法分离出生10~15d的大鼠海马神经元,从形态学及电生理学方面鉴定细胞活性。结果表明,该法可获得形态和生理特性良好的单个海马神经元。用0.5μmol/L的TTX阻断Na+通道后,分别记录到典型的全细胞电压依赖性钾离子通道电流以及延迟整流钾通道电流。证实该方法适用于海马神经元膜片钳研究,对深入探讨药物和毒物对海马离子通道及信号转导机制的作用有重要价值。

Abstract: The hippocampal neurons were isolated by enzymatic and mechanical methods from 10-15 day postnatal rat, and its activity were verified by morphology and electrophysiology. Results showed that single hippocampal neuron with good shape and physiological characteristic can be obtained by using this method. After blocked Na+channels by 0.5μmol/L TTX, standard whole cell voltage dependent potassium current and delayed rectifier potassium current were recorded. This proved that the method is suitable for study on hippocampal neurons by patch clamp,and it have important value for deeply research the effects of drugs and toxins on hippocampal ion channels and on its signal transduc-tion mechanism.
