非清髓性异基因骨髓移植小鼠模型的建立

Establishment of Mouse Model of Nonmyeloablative Bone Marrow Transplantation

摘要: 为探讨非清髓造血干细胞移植小鼠模型的制备,以10~12周龄C57BL/6(H-2b)雌性小鼠为受体,以10~12周龄BALB/c(H-2d)雄性小鼠为供体,移植前采用环磷酰胺200mg/kg)加5 Gy γ射线全身照射的非清髓预处理方案,移植后36 d检测雌性受体小鼠体内Y染色体特异性基因表达作为移植植活指标。结果发现,非清髓的预处理方案下可以实现异基因造血细胞的植活同时发现移植回输的造血细胞数量对于造模成功率有着关键作用,在回输异基因骨髓细胞数量为2×107个时移植植活率为80%(8/10)。

Abstract: To establish a mouse model of nonmyeloablative bone marrow transplantation, we used fe-male C57BL/6 (H-2b) mice as recipients and male BALB/c (H-2d) mice as donors.The re-cipients received Cyclophosphamide (200mg/kg) and total body irradiation (5 Gy).This regime is nonmyeloablative On the 36th day after transplantation,we detected the Y chromosome specific gene in bone marrow cells of recipients mice. We found that engraftment of allogeneic hematopoietic stem cells can be established under the nonmyeloablative pretreatment regime.We also found that the number of hematopoietic stem cells is very important for the success rate of making model.The rate of engraftment is 80% (8/10) when the recipients mice were given 2 × 107 bone marrow cells.
