小型猪心肌梗死模型的制作和判定

Establishment and Identification of Minipig Model with Myocardial Infarction and ischemia

摘要: 制作小型猪心肌梗死和缺血模型,并建立活体判断模型建成与否的观察指标。方法:12只16~22kg小型猪分为2组,每组6只,第一组结扎回旋支制作心肌梗死模型,第二组用ameroid环制作心肌缺血模型。用心肌99mTc-MIBI血流灌注显像在活体确定其梗死区或缺血区,并在处死动物后用TTC法验证其梗死或缺血灶。结果:成功建立小型猪心肌梗死和缺血的动物模型,99mTc-MIBI心肌血流灌注显像可在活体上显示小型猪模型心肌梗死灶的放射性缺损区和缺血灶灌注缺损区的放射性再充填。离体心肌TTC染色可显示相应的梗死或缺血灶。结论:结扎回旋支和ameroid环缩窄回旋支是制备小型猪心肌梗死和缺血模型的较好方法,可用99mTc-MIBI心肌灌注显像在活体上确定模型成与否。

Abstract: To establish the minipig model with the myocardial infarction or ischimia,and observation marker proving the model. 12 minipigs with body weight 16-22 kilogram were divided into two groups,one was treated with circumflex branch ligation for the myocardial infarction,the other was treated circumflex branch with ameroid constrictor for myocardial ischemia. Both the models underwent exercise and resting myocardial perfusion imaging with single photon emission computed tomography (SPECT) using 99mTc-MIBI,they also underwent the staining with 2,3-triphenyltetretrazolium chloride in order to distinguish the myocardial infarction or ischemia with isolated heart. The results showed that the left ventricle of the model was observed the defect of 99mTc-MIBI in the infarction region where the staining of TTC was no uptake. The other model was observed rarefaction of 99mTc-MIBI in the ischemia region where TTC was less uptake.The minipig with circumflex branch ligation was the better model for the myocardial infarction and circumflcx branch constriction for ischemia. 99mTc-MIBI myocardial perfusion can define the infarction or ischemia segment in the living body while the TTC can also define in the isolated heart.
