软骨细胞三种凋亡模型的建立

Establishment of Three Models on Chondrocyte Apoptosis

摘要: 为了观察药物对骨关节炎中软骨细胞凋亡的影响,建立了软骨细胞三种凋亡模型。将硝普钠(SNP)、全反式维甲酸(ATRA)、肿瘤坏死因子-α(TNF-α)分别加入软骨细胞培养体系,建立三种不同药物诱导的软骨细胞凋亡模型,采用Annexin v/PI双参数法以及透射电镜对软骨细胞凋亡进行定量、定性检测。在兔软骨细胞培养体系中分别加入2mmol/L SNP,10-4mol/L ATRA 24h后,在人胚胎软骨细胞培养体系中加入30μg/L人TNF-α 24h后,均可成功建立软骨细胞凋亡模型。提示硝普钠、全反式维甲酸、肿瘤坏死因子-α加入软骨细胞培养体系中,可建立软骨细胞凋亡模型。

Abstract: To investingate the effect of drug on apoptosis of chondrocyte in osteoarthritis, the three chondrocyte apoptosis models were established,treated with sodium nitroprusside (SNP)、all-trans retinoic acid (ATRA)and tumor necrosis factor-a (TNF-a) respectively. The apoptosis rate were analysed by Annexin V/PI with flow cytometry and the characteristic alterations of morphology were detected by electron miscroscope. When rabbit chondrocyte were incubated with SNP (2mmol/l),and ATRA (10-4 mmol/l),and chondrocyte coming from fetal were incubated with TNF-a(30/μg/L) for 24h,they all could induce apoptosis in chondrocyte in vitro. The re-sults suggested that the chondrocyte apoptosis models could be established by sodium nitroprus-side、all-trans retinoic acid and tumor necrosis factor-α respectively.
