摘要: 利用人黑色素瘤scid小鼠皮下移植实验中偶尔发现的肺转移瘤组织,接种至scid小鼠皮下,经体内反复筛选,成功建立了一株人黑色素瘤高转移模型SCI-375,至今在体内筛选2年共21代,表现为移植瘤局部侵润、100%自发肺转移合并79%腋下淋巴结转移,组织病理学观察和染色体分析表明,该模型保持了人黑色素瘤的基本特征。同时,利用筛选的肿瘤组织在体外初步建立了一个相应的细胞系,经体内移植实验证实,细胞系保持了原模型的高转移特性。本模型的建立,为研究肿瘤的转移机理和抗转移药物的筛选提供了一个理想模型。

Abstract: We have successfully established a highly metastatic model of human melanoma in scid mice (SCI-375). The selection of metastasis was obtained by using successively subcutaneous implantation of lungs metastatic tissue in scid mice. The metastatic rates were 100%and 80% respectively in lungs and lymph node from 6-21 passages. The lung metastatic tumors were similar to the original human melanoma in histopathological features and chromosomal karyotype. This metastasis model may be used as a tool for the research on the metastatic mechanism and treatment of the human tumor.
