犬细小病毒的分离与鉴定

摘要: 应用猫肾F-81系传代细胞,采用同步接毒的方法及无牛血清细胞培养法,从临床患出血性肠炎大粪中分离到一株病毒。通过对第6代细胞培养病毒液进行电镜形态观察、病毒理化特性试验、核酸型測定、幼犬人工感染试验、血凝及血凝抑制试验,以及用标准抗CPV免疫清进行病毒中和试验及免疫电镜观察等系统鉴定,证明该病毒株为犬细小.病毒。在病毒的分离培养过程中,比较了细胞培养维持液中牛血清成份对病毒增殖的影响,发现含2%胎牛血清的细胞維持液对犬細小病毒的生长有抑制作用。采用无牛血清的细胞培养维持液,则有利于犬细小病毒的增殖。建议采用无牛血清细胞維持液培养犬细小病毒,以利提高犬細小病毒的产率。

Abstract: "A strain of virus, isolated from a stool specimen of a canine with clinical hemorrhagic enteritis, was confirmed to be Canine Parvovirus by its physicochemical characteristics, analysis of its nuclear acid type, electronic and immunoelcctronic microscopic observation, experimental infection on puppies, hemagglutination test and hemagglutin-ation-inhibition test, and serum neutralization assay. It was found that no fetal bovine serum in the maintaining fluid of the cell culture and inoculation on fresh cells were beneficial to the propagation of this virus. "
