    Establishment of Bronchiectasis Model in Rat

    To observe the law of bronchiectasis and study the clinical pharmacology, sig-nificance of the result of antibiotic and Chinese traditional medicine.A bronchiectasis animal model was established by ligating the rat’s bronchus and then injecting ps. Aeruginosa into the bronchial lumen,another one was established by injecting ps,aeru-uginosa directly into bronchial lumen without ligation. The results showed that both models were similar to human brochiectasis in histopathology,volume of bronchial secretion and the amount of ps, aeruginosa in bronchial lavage. It was concluded that both model was available for the study of bronchiectasis.

    SD rats were intraperitoneally injected with ovabulmin.Two weeks later,the rats were intravenously challenged with ovalbumin,and the airway resistance,the pulmonary compliance were detected by breathe fluent analysis software. The results showed that after challenging with ovalbumin,airway resistance of the rats were significantly increased,and pulmonary compliance was obviously decreased after the challenge.The decrease of the pulmonary compliance was significantly correlated with the increment of the airway resistance.The results suggested that the allergic asthma model was successfully established. It is useful for researches on non-glucocorticoids immunosuppressive for asthma therapy,as well as the mechanisms of glucocorticoids treatment in allergic asthma.

    The authors had isolated skeletal muscle satellite cells by mechanic and enzymic digestion,purified them by Pecoll discontinuous density gradient centrifugation and identified them immunocytochemically by Desmin-labeling,and multiplied the satellite cells by administration of promoting heptocyte growth factor (pHGF). The purity of harvested satellite cells exceeded 90%.It was suggested that the pHGF was usful for prompting satellite cell growth by shortening the lag period and enhancing their proliferation capacities.

    To investigate the regulatory role of osteogenic growth peptide (OGP) in rat osteoblast-like cell.The isolated osteoblastic cells from calvaria bone of newborn rats were cultured in vitro. They were divided into two groups,one was added with OGP,the other was added with DMEM as the control. The medium was changed twice a week. On the 3rd、7th、10th、14th、21th culture-day,the activity of alkaline phosphatase、acid phosphatase,the contents of proteoglycan、collagen and Ca in the cells and extracellular-matrix were detected. The results showed that,the alkaline phosphatase activities and calcium level in the osteoblastic cells of OGP group on the3rd,7th, 10th,14th, 21th culture-day were significantly higher than those of the control groups (P<0.01-0.05), the collagen level of OGP group were 1.5 times higher than the control (P<0.01).It was concluded that OGP can greatly promote the proliferation of osteoblast-like cells and produce more bone tissues.
    Pathological Observation on Wound Healing of Deep Partial Thickness Burn in Rats by Scalding with Different Length of Time

    To explore the mechanisms in wound-healing of deep partial thickness burns of skin, an animal model was prepared by scalding SD rat skin with water at 80℃ for 6 and 15 seconds in group 1 and 2 respectively,and then followed by light microscopic ob-servation on the natural healing of the wound at fixed intervals. The local microcircula-tory impairement of the rats in group 1 was found less serious,7 days after the scald proliferation of the epithelium of hair follicles,which remained deep in the corium,took place and covering the wound surfaces, and 14 days after the scald the wounds were almost healed. In group 2,the microcirculatory impairement was so obvious.Seven 7 days after scald the wound showedwere deepening progressively from deep partial to full thickness burns,another week later abscess formed beneath the scabs that hindered the wound from healing.It is concluded that the degree of impairement of microcirculation is highly correlated with the length of time of scalding. The severity of microcirculatory impairement and the inflammatory reaction beneath the scabs are both major factors that affect the wound healing.
    Study on Latent Infection of Mousepox

    The conventional and clean KM,BALB/c and C57BL/6J mice were detected for LCM( lymphocytic choriomeningitis virus), EHFV( epidemic hemorrhagic fever virus),Mouse pox, MPV( mouse hepatitis virus), Sendai virus infetion by using IFA,ELISA,ABC( avidin-biotin-peroxidse complex), and electron microscope before and 15,30,60 days after undergoing animal experiments.The results revealed that 60 days after the animal experiments, ectromelia virus was found in the liver,mesenteric lym-phaden,Payer’s patches, and spleen of seronegative(ELISA,IFA ) KM and BALB/c mice,while in the other groups no ectromelia virus infection were found.The findings suggest that the mice with latent infection of ectromelia virus,despite of sero-negative,would give rise to an outbreak when they were under a long stress of animal experiment ,and the viral antigen detection of Mousepox should be used in routine microbio-logical monitoring.

    Two different types of artery canulation were established in piglets to study the time consumng blood losing,changing rate of body temperature,vital organ perfusion during canulation and deep hypothermia cardiopulmonary bypass. The results showed that the time consuming,blood losing in femoral artery canulating group were less than those in ascending aorta canulating group,and there were no differences between two groups in changing rate of body temperature, and vital organ perfusion under 80ml·kg-1·min-1 flow rate. Established cardiopulmonary bypass using modified femoral artery canulating method is a relatively simple mode,it appeared feasible to preserve thoracic-cardiac stability.

    The Primary Contents analysis(PCA) of the scapula were carried out in M.mulatta distributing over Taihang mountains (Henan). The variables of the scapula were compared with those of monkeys of the same species living in other regions in China (Sichuan, Yunnan,Hubei and Hunan provinces).The results suggested that the scapular morphology of M. mulatta in Henan is significantly different from those race living in other regions. The morphological differences suggested that the scapular features play an important role in differentiating the subspecies and locomotor behavior of M. Mulatta in China.
    A New Orthotopic Transplantation Technique for Establishing Human Colon Cancer SW1116 Model in Nude Mice

    To establish a human colon cancer model in nude mice,a new anastomotic orthotopic transplatation technique was developed. Human colon cancer SW1116 cells were implanted subcutaneosly and passaged in nude mice for 4 generations,no metastasis was found. From 5th generation the tumors were resected subtotally when reaching 1.5 cm in diameter. After the tumor grew again, 1 of the 8 mice appeared metastatic lymph node in the armpit of the forelimb. A medical adhesive,OB Glue,was used in orthotopic transplantation of the intact SW1116 tumors from the metastatic lymph node.At the first 4 passages the metastatic rate was low. From the 5th passage, the spleen of Vmice were also resected at the time of tumor orthotopic transplantation with which the metastatic rate was high. From 6th to 10th passage the metastatic rate was 100% (40/40) to the mesenteric lymph nodes, 75%(30/40) to the livers. It can be considered that the metastatic SW1116 sub-population (subpop) was obtained.The orthotopic Transplantation method and SW1116 subpop model of nude mice may be useful for further study of human colon cancer.
