A New Orthotopic Transplantation Technique for Establishing Human Colon Cancer SW1116 Model in Nude Mice

Abstract: To establish a human colon cancer model in nude mice,a new anastomotic orthotopic transplatation technique was developed. Human colon cancer SW1116 cells were implanted subcutaneosly and passaged in nude mice for 4 generations,no metastasis was found. From 5th generation the tumors were resected subtotally when reaching 1.5 cm in diameter. After the tumor grew again, 1 of the 8 mice appeared metastatic lymph node in the armpit of the forelimb. A medical adhesive,OB Glue,was used in orthotopic transplantation of the intact SW1116 tumors from the metastatic lymph node.At the first 4 passages the metastatic rate was low. From the 5th passage, the spleen of Vmice were also resected at the time of tumor orthotopic transplantation with which the metastatic rate was high. From 6th to 10th passage the metastatic rate was 100% (40/40) to the mesenteric lymph nodes, 75%(30/40) to the livers. It can be considered that the metastatic SW1116 sub-population (subpop) was obtained.The orthotopic Transplantation method and SW1116 subpop model of nude mice may be useful for further study of human colon cancer.
