Experimental Study on Rabbit Atherosclerotic Plaque Rupture Model

To establish atherosclerotic animal model whose plaque could be evoked into rupture, male New Zealand white rabbits were fed with a high-cholesterol diet for 2 weeks, then underwent balloon-induced arterial wall injury, and fed with a high-cholesterol diet for 6 weeks and a regular diet for 8 weeks. Plaque rupture and thrombosis were triggered by injection of Russell's viper venom (0.12 mg/ kg,ip) and histamine (0.016 mg/kg,iv). The area of thrombosis on atherosclerotic aorta, morphological character of plaque rupture, levels of blood lipid and lipoproteins were determined by image analysis, electron and light microscope, chromatometry, respectively. The results showed that the surface area covered by thrombus was 0.9±1.1 mm
2
in normal group and 77.7± 53.8 mm
2
in model group.Microscopy of arterial samples from model group showed obvious ulcers occurred and inflammatory cells infiltrated in shoulder area of plaque.Macrophages and lipid were rich in plaque. Content of cholesterol in abdominal aorta was 6.6 ±1.4 mg/g wet tissue in normal group and 24.2±7.7 mg/g wet tissue in model group. Conclusions:This animal model mimicked atherosclerotic plaque rupture and thrombosis,and could be used in study on mechanism of plaque rupture and screening drugs of plaque stabiligation out relational compound.

To establish the allergic asthma model, guinea pigs were intraperitoneally injected with ovabulmin, and challenged by inhaling ovabulmin with ultrasonically nebulised inhaler two weeks later. The total number of cell and eosinphil count in brochial alveolar lavage fluid (BALF), eosinphil count in plasma, airway resistance,pathology were detected. The results showed that eosinphil count in plasma and BALF of the model animal were significantly increased after the challenge compared with the control, intrapul-monary pressure were increased after inhaling histamine, and eosinphil imflammion were found in patho-logical exanination.

To establish the neonatal Mongolian gerbil model of lead poisoning, eleven litters of 6-7days neonatal Mongolian gerbil were randomly divided into normal,low and high lead group. The animals were injected intraperitoneally with 0.9% saline or lead acetate trihydrate at the doses of 12,36 mg/kg (according to bodyweight) every other day and were breastfeeding by their mother.The weight of body and brain,lead density of blood and haemoglobin of neonatal Mongolian gerbil were measured. The pathological changes of kidney and brain were observed. The results showed thatthe weight of body,brain, and haemoglobin in low-lead and high-lead group were remarkably decreased (

It is suggested that the method for establishment of lead poisoning model of neonatal Mongolian gerbil by intraperitoneal injection was a kind of simple,stable and controllable way, and the model may be used for study of early stage nervous system and blood vessel system.

To establish monitoring methods of biochemical markers in Meriones Unguiculataus,the biochemical markers of Meriones Unguiculataus were screened,and its electrophoretic conditions of isoenzymes and proteins were probed according to cellulose acetate membrane electrophoresis of proteins and isoenzymes. The polymorphism of 9 microsatellite loci was detected in Z:ZCLA Mongolian gerbil from Zhejiang center of laboratory animal , Zhejiang Academy of Medical Sciences. The results showed that 8 loci were polymorphic and locus 1 had 1 allele, and others had 2.4 alleles. The average of alleles is 2.6.

In order to detect rodent coronavirus, a pair of PCR primers were designed according to the compared sequences of rodent coronavirus and SARS coronavirus. The primers derived from the specific and conservative N gene,RT-PCR method was established. Result of rodent RT-PCR amplification showed the amplified products of 148bp length; RT-PCR method of rodent coronavirus has been used to detect the samples. The rodent RT-PCR method is important to determine early rodent coronavirus and discriminate SARS coronavirus.

In the present paper ,11 variables of mandibles of adult male mice from three strains were measured. The mice involved were 16 KM,10 BALB/c, 10 ICR respectively.The variables of mandibles were analyzed by Enter Independents Together and Use Stepwise Method of SPSS for Windows 10.0. These variables were used to establish discriminant functions for distinguishing among the three differ-ent strains. The correct discriminant rates both reach 100% by original grouped cases;the correct discriminant rates both reach 97.2% by cross-validated grouped cases.

To study the effect of coronary stent implantation on the coagulation and platelet function of rabbits, eighteen New Zealand white male rabbits were underwent precutaneous transluminal coronary stenting. Their blood samples were collected before and at 1,2,4,24 hour after the stent implantation, respectively. The coagulant parameters of thrombin time (TT), prothrombin time (PT), activated partial thromboplastin time (APTT) and the fibrinogen, P-Selectin and thromboxane B

) level in plasma, platelet number, and tested platelet aggregation function were detected. The coagulation parameters of TT, PT, APTT had no significant change after the stent implantation, but the platelet number and the fibrinogen level in plasma was decrease after stent implantation and came back to the normal level in the 24 hour after the surgery. The level of platelet aggregation rate and P-Selectin and TxB

in plasma was higher after the stent implantation. Therefore, the coronary stent implantation had significant effect on the platelet activation,thrombosis and the activation of congulant system, It was approved that it is necessary to inspect the platelet activation and therapy with antithrombotic medicine after the stent implantation simultaneously.
