    Biological Properties of Orthotopic Implant Models for Human Gastric Carcinoma in Nude and SCID Mice

    Objective To explore differential biological properties of local and metastatic human gastric cancer after orthotopic inplantation in nude mice and SCID mice. Methods MKN-45 human gastric carcinoma cell line was inoculated subcutaneously in nude mice to develop s.c. growing tumors. Orthotopic implant model simulating human gastric cancer was established by suturing histologically intact human tumor tissue on the stomach walls of nude mice and SCID mice. Subsequently, we observed the take rate of the orthotopic tumors and their biological properties of tumor growth and metastasis. Results ① 100% tumor take rates were observed in both strains of mice.② The tumor volume in the stomach of nude mice was 2884±1337 mm3,the metastases of local and distant lymph nodes were noted in 67% of the mice, the rates of liver, lung and diaphragm metastases were 83%, 33%, 8%, respectively.③ The tumor volume in the stomach of SCID mice was 4582±1326 mm3P<0.05).The incidences of liver metastases in SCID mice were almost similar to those of nude mice which were 90% (P>0.05). In contrast, there was a significant increase in the incidences of lung and diaphragm metastases, which were 100% (P<0.01) and 60% (P<0.05),respectively. Conclusions The results suggest that SCID mice are more suitable for establishment of orthotopic implant models for human gastric carcinoma compared with nude mice.

    Objective To explore effects of PASG knockout on brain development and lay a foundation for research on molecular etiology of glioma. Methods No. 10-12 exons of murine PASG gene were selectively deleted with classical knockout method. Histological analysis and western blot assay were employed to examine CNS cell morphology and gene expression in mutant mice. Results PASG mutant mice displayed functional abnormality in multi-organs (brain, lung, kidney, bone and so on), especially neurological development retardation, histological changes in hippocampal region and expression changes of related genes. Conclusions PASG is essential for regulating embryo development and cell proliferation. The disruption of PASG caused abnormality structurally and functionally in organs of mice. Moreover, the close relationship between neural stem cells and cells in hippocampus provided a new access to molecular mechanism of glioma formation.
    Establish Chronic Renal Failure Model with High-grade Proteinuria in Rat

    Objective: To establish Chronic Renal Failure (CRF) with high-grade proteinuria rat model. Methods: The 5/6 nephrectomy rat was replicated and fed with high-protein diet. Results: Volume of Scr and 24h proteinuria in CRF rat’s with 40% high-protein diet group were increased significantly than those of normal diet control and 20% high-protein diet group. Renal pathology showed more severely neprosclerosis and matrix fibrosis in 40% high-protein diet group than other groups. Conclusion:CRF with high-grade proteinuria rat models can be established by supplying high-protein diet to replicated 5/6 nephrectomy rats.

    Objective To investigate the dynamic changes of renal pathological morphology in the rat model with 5/6 nephrectomy. Methods Observed by light and electron microscope. Results The characteristics of renal dynamic changes of 5/6 nephrectomy rat:glomenrulus enlarged ,mesangial cells hyperplasia and tubule expansion at 2 weeks. At 8 weeks, mesangial cells severe hyperplasia and tubule-interstitial inflammation and expansion.At 12 weeks,focal segmental glomerular sclerosis and renal interstitial fibrosis. At the last of 40 weeks, we could see most of glomerulus fibrosis and sclerosis and tubule-interstitial severe fibrosis. Conclusion The changes of 5/6 nephrectomy in rats were accorded with renal fibrosis in human, it was a ideal model to research in chronic renal failure and renal interstitial fibrosis.

    Objective To study the effect of ethanol extract from Qufeng tongluo recipe on cerebral blood flow (CBF) in rats with acute cerebral ischemic injury. Methods Using a modified MCAO method, the authors established the model and flushed the ischemic territory via ICA with Chinese recipe solutions (Qftl) at the early start of reperfusion that 2 hours after MCAO. Rats of control (ip group) were ip 3 ml Chinese recipe solutions at the right time. Results Compared to model group, CBF of Qftl and ip group was increased significantly (P<0.01,P<0.05 respectively) and neurological deficit scores were increased (P<0.01,P<0.05 respectively). The CBF and neurological deficit score was significantly different between Qftl and ip group (P<0.05). Conclusion (1) Focal flushing via ICA is an effective method. The mechanism of this treatment is not only about blood dilution and capacity increasing, but about focal effects of different medicines.

    Objective To establish rabbit iliac arterial re-stcnotic models. Methods Endothelium-denudation in iliac artery was conducted by Fogarty catheter and second injury by balloon angioplasty, and thirty-six New Zealand White rabbits were used in the experiment. Saphenous artery was accessed for the first injury,and femoral artery for the second injury. Arteriography, electroscopic and pathological examinations were used to detect rc-stcnotic samples excised immediately and to day 84 after balloon angioplasty. Results Among the 34 limbs which were successfully done, re-stenotic rate was 50%~80%) in 28 right iliac arteries, and less than 50% in 6. Main components of neointima and media were vascular smooth muscle cells (VSMCs). With the progress of the time after operation, numbers of VSMCs were decreased while the foam cells were increased, ccllular components were decreased while the collagen components were increased, proportion of neo-intima was increased while that of media was decreased. Conclusion The improved rabbit re-stenotic models were stable and reliable, and the surgical procedures were easily as well.

    Telomeres are special DNA-protein complexes, which situated in the ends of eukaryotic chromosomes. Telomerase is a specialized ribonucleoprotein complex with reverse transcriptase activity that is capable of extending chromosome ends with specific telomeric DNA sequences by using a portion of its RNA component as a template and using 3’ telomere DNA as primers. It consists of two subunits TERC and TERT. The mtere-/- mice is one of the ideal model for study the functions of subunit TERC and related diseases. This article reviewed the applications of the model on study of tumor, aging, blood and reproduction.

    Vascular dementia (VD) is a syndrome manifested by declines in cognitive function which is the results of cerebral blood flow disorders and functional disorders caused by a series of cerebral vascular factors. It is of vital importance to establish a VD animal model caused by cerebral ischemia to deeply explore the etiology, pathological features of the disease and to insure an early diagnosis, treatment and even prevention for the patients. In this study, a review was performed in the research progress of experimental VD animal model.
