    To establish a method of isolating and obtaining porcine bone marrow mesenchymal stem cells (MSCs)in vitro,and to assess the possibility of MSCs as a new ccll source for tissue engineering. Three ml bone marrow was extracted from the posterior superior iliac spine of the mini-pig and culturd by 1×106/ml cells. Some cells were attached to the culture dish after the second day.The adherent cells approached confluence 10 days after plating and then every passage was cultured for 5~7 days. The primary culture and 10th passaged adherent cells were induced to differentiate into osteoblast and myogenic cells. The primary culture and the passaged cells were spindle-shaped in appearance.They both might be induced to differentiate into bone marrow mesenchymal cells and osteoblast and myogenic cells under special environment. The method for isolating and culturing porcine MSCs was feasible.
    Establishment and Observation of Gentamicin-induced Acute Tubular Necrosis in Rat

    Acute tubular necrosis animal models were established by subcutaneous injection of gentamicin(GM) 400 mg/kg/day for 2 days in adult Wistar rats and the effects of recombinant human epidermal growth factor (rhEGF) on the renal function and histology were observed. On 5th day after the first GM exposure,epithelial cells of proximal tubules were diffusely necrotic and then gradually recovered to normal on 21 th day. Renal function values of EGF-treated animals returned to normal 7 days earlier than that of GM-treated animals. At the early stage of renal injury,the histological changes were not different from each other. When examined at days 11 and 15,kidneys from rats that received rhEGF were improved in histopahtological appearances compared with those from GM-treated animals. These results suggests that the rhEGF enhances re-nal tubular repair and accelerates the recovery of renal function in a model of gentamicin nepro-toxicity in rats,and the effects of rhEGF are related with a certain degree of injury of the models and its dosage.

    The blood pressure (BP),heart rate, body weight and organ weight related to hypertension were monitored and detected in SHR through five generations,the results showed that the SHR had formed hypertensive characters,the average BP in adult SHR was 189±6 mmHg nearly equal to the original SHR (193 mmHg).From 6th week the hypertensive character was appeared in SHR and the BP increased gradually thereafter,reached highest level on 25th week in male (200±9mmHg) and 17th week in female (178±7mmHg).The BP levels of SHR in different ages were significantly higher than that of the background strain (WKY),whereas the body weight (BW) were lower than WKY,about 86% of WKY on birth and 67% at the age of 100 days .The organ coefficient of the brain and kidney were higher than that of WKY. Both the weight and organ coefficient of the heart and hypophysis were significantly higher than those of WKY .In period of disease development,SHR also appeared typical pathological changes in cardiovascular system. These pathological changes could cause heart hypertrophy and combining symptoms in heart, brain and kidney.

    Objective The purpose of this study is to provide animal model of diabetes Ⅱ and basic data for studying the development principle of the diabetes and diabetic complication. Methods:The rat model of diabetes Ⅱ was made by fed with a diet enriched with energy and ip low-dose streptozotocin (STZ 30mg/kg). The indexes related to the development of diabetic complication were detected. Results:The blood glucose level of treated rats was significantly higher than that of normal control rats,and the sensitivity to insulin was decreased. The level of advanced glyco-sylation end products-peptide (AGE-P) and lipoid peroxidation products malondialdehyde (MDA) in serum and the activity of aldose reductase(AR) in kidney of the diabetes model rats were remarkably increased, compared with whose of the control rats,whereas the superoxide dis-mutase (SOD) activity in serum of the models was significantly decreased. Conclusion:The diabetes Ⅱ rat model can be successfully established by fed with a diet enriched with energy and ip low-dose streptozotocin. The levels of indexes related to diabetic complication were changed. And this change may induce the development of diabetic complication.

    To study the hemotopoiesis capabitily supported by myeloid matrix cell in the liquid culture medium,the mice myelid kayocyte culture in vitro, after formation of sticking-wall layer of matrix cell in the liquid culture medium, changed culture solution and then replant myeloid karyocyte. Results showed that the cell colony was found on the stickingwall matrix cell. Morphologically, they can be classified into two groups, matrix cell colony and hematopoietic cell colony. The former could be seen occassionally while the latter appeared in great number. The conclusions are that the formation of hematopoietic cell colony require the micro-environment of myeloid matrix cells contact,and has a certain retroaction on the growth and development of matrix cell. The study created a liquid cultivation of hematopoietic cell colony.

    To study the difference between high dose sodium-chloride diet associate with high dose N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) gavage or low dose MNNG in drinking water in inducing gastric carcinomas in rats. Sixty-six male 6-week-old Wistar rats were divide into 3 groups. Group 1 rats were given MNNG in drinking water at a concentration of 100 mg/L for 17 weeks and were simultaneously fed a diet supplemented with 8% NaCl for 20 weeks. Group 2 rats were initially given MNNG (200 mg/kg b. w. ) by gavage at days 0 and 14 as well as saturated sodium chloride solution (S-NaCl,1 ml per rat) every 3 days during first 3 weeks (six times), and then treat with high dose of sodium-chloride diet for 17 weeks. Then these two groups of rats were treated with normal diet for 15 weeks. Group 3 rats were fed on normal diet for 35 weeks. At week 20,ten rats of group 1 were killed and at the end of week 35 all surviving animals were killed. All the specimens were histopathologically investigated. The result showed that the incidence of glandular stomach adenocarcinoma in group 1 was 20% in week 20 and in week 35 was 90%. The incidence of forestomach cancer in group 2 was 68.4%. These data indicated that the method of high dose sodium-chloride diet associate with low dose MNNG in drinking water is more effective in inducing glandular stomach adenocarcinoma.

    Objective:“Caihe No. Ⅰ、Caihe No.Ⅱ”is a preparation made from pure herbal medicine and has been using for frosbite treatment in human for a long time. Its therapeutic effect on rabbit was evaluated here. Methods : Silver dollar was frozen by liquid nitrogen (-196℃) and sticked closely onto the skin of the rabbit to establish grade Ⅱ frostbite animal model. The therapeutic effect of “Caihe No. Ⅰ、Ⅱ”was analyzed with the control group. Results:① Pathological changes occurred for Ⅱ degree to deep Ⅱ degree at 5s after frostbite.② After 4 day-period treatment of “Caihe No. Ⅰ、Ⅱ,I degree frostbite were healed. There was significant difference between “Caihe No. I、Ⅱ” and the control group. Conclusion: “Caihe No. Ⅰ、Ⅱ ” have therapeutic effect for frostbite in rabbit. “Caihe No. Ⅰ”is a decoction,while “Caihe No. Ⅱ”is a cream, and the latter is more convenient for pernio in winter. The results of the experiment is same as the clinical result, so its curative effect is reliable.

    The adult Macaca mulatta from the Taihang Mountains had been measured about seventeen variables of the mandibles (male 9,femal 16). Employing SPSS for windows 10.0,ANO-VA test showed that only three variables are significant differences between sexes. Sexual discriminant functions were constructed by means of Enter independents together and Use stepwise method. The results showed: the discriminant rate of the former is 100%,and the latter is 88.0% . But the latter is simpler.

    Diabetic mellitus animal models include spontaneous and induced rodent or non-human primate etc. Some of the characteristics of these animal models, such as disease development、changes of histopathology are similar to those of human diabetes mellitus. Diabetic mellitus animal models can be applied to research of preventive medicine、etiology and mechanism of human diabetes and its complications, and also to the exploiting of new medicines for diabetes. With the development of new biotechnology,diabetic mellitus animal models will play a more important role in conquering the disease of diabetes mellitus.
