    Establishment and Assessment of a Mouse Model of Post-infarct Cardiac Rupture

    Objective To establish and assess a mouse model of post-infarct cardiac rupture to characterize pathogenesis of ventricular rupture. Methods One hundred and forty male C57BL mice were randomized into 4 groups - sham operated,high, middle and low level ligation group based on location of occlusion left coronary artery and underwent open-chest surgery to induce myocardial infarction (MI). The time-window,location and incidence of ventricular rupture were observed during 7 days postinfarct.Hemodynamics, gross and histological examinations were undertaken after 7 days following MI. Results The time-window of left ventricular rupture was at 2-6 days after MI.Incidence of rupture in mice of middle level ligation group was significantly high than other groups (P<0.05). The infarct size of high level ligation group was significantly high than other groups (P<0.05). The high ligation group mice had worse hemodynamics compared with SH and other groups(P<0.05).The pathological examination indicated that score of intramural hemorrhage and accumulation of inflammatory cells in middle ligation group was significantly high than other groups (P<0.05). Conclusions The mouse post-infarct cardiac rupture model were stable and reliable, and may be used for experiment research of post-infarct ventricular rupture.
    Establishment of Endometriosis Model in Rats with Intact Uterine Tissues

    Objective To establish the endometriosis models in rats. Methods Twenty-six mature female SD rats were used. The rat endometriosis model was established by autotransplantation with uterine tissues. Eight weeks after the autotransplantation,the second laparotomy was performed for observing the size,conglutination and pathological changes of the autotransplants. Results Saccate autotransplants were found in twenty-one rats, all of which were embedded in the peripheral tissues. The morphology and structure of most ectopic endometrium were as same as the eutopic endometrium, though there were less epithelium and stroma in ectopic tissues. However, in a few cases, there was either epithelium or stroma. Conclusion The endometriosis model established is successful, which can provide an ideal model for further study of the mechanism and medical treatment of endometriosis.
    Biological Characteristics of BNX Mice and its Preliminary Application on Tumor Model

    Objective To supply some basic data for extension and application of BNX mice through observation on generally biological properties and peculiarity of tumor growth and metastasis after tumor implantation. Methods By the method of periodically measuring the weight、analyzing breeding character, graphing growth curve、examining hematological and blood biochemical parameters,we explored biological characteristics of BNX mice. At the same time,we also established tumor animal models of human gastric tumor (MKN-45) and human melanoma (SCI-375) in BNX mice through orthotopic and subcutaneous tumor implantation. Results It showed that there was a statistically significant difference of body weight between male and female BNX mice within 4?30 weeks (P<0.05). The number of WBC in peripheral blood haematology of BNX mice was only equal to twenty-five percent of that of normal mice,while no statistically significant differences in biochemical parameters was observed between BNX mice and normal mice. The metastatic rates of orthotopic implant models of human gastric tumor (MKN-45) and subcutaneous implant models of human melanoma (SCI-375) in BNX mice were much higher than in nude mice and SCID mice.Conclusions These results suggest that BNX mice are more suitable for the study of establishment of tumor metastatic models compared with nude mice and SCID mice.
    Establishment of Rabbit Model of Allergic Rhinitis and Histological Observation of Nasal Mucosa

    Objective To establish a credible and convenient rabbit model of allergic rhinitis and study the histopathological and immunohistochemistrical change of nasal mucosa. Methods Both sides of nasal cavity in rabbits were instilled with 0.1ml 10% toluene -2,4-diisocyanate (TDI) olive oil solution one time every day for continuous 7 days;two days after the last instillation, provoke allergic reaction in rabbits through the above mentioned instillation method for 15 times and record the symptoms and physical sign respectively;execute the rabbits day after the last provocation and examine the nasal mucosa for histopathological and immunohistochemistrical changes. Results With the sensitization of TDI, typical allergic rhinitis symptoms and physical sign were observed in experimental rabbits. Patho-logical examination of nasal mucosa showed obvious edema and vasodilation;scattered or focal infiltrations of inflammatory cells composed of eosinophils mainly were visible on lamina propria;substance P expressed and increased drastically, staining intensified. Conclusion Establishment of rabbit model of allergic rhinitis by instillation with 0.1ml 10% TDI olive oil solution into both sides of the nasal cavity is proved simple, reliable, and with high success ratio and low mortality. The physical sign, pathological changes and immunological reaction of substance P of rabbit model are conparable to clinical symptoms and pathological changes;this model may be used for the study of allergic rhinitis and drug screening.

    Objective To study the effects of Lithium chloride(LiCl) on inhibition of the proliferation of human SMMC-7721 cells and the growth of Tumor in hepatocarcinoma-bearing mice. Methods Using MTT assay to detect anti- proliferation effect of Lithium chloride on SMMC-7721 cells, applying Flow cytometry(FCM) to observe whether Lithium chloride can induce apoptosis in SMMC-7721 cells or not;mouse hepatocellular carcinoma cells H22 were subcutaneously injected into KM mice to establish animal model of hepatocarcinoma-bearing mice,and LiCl were administered to the tumor-bearing mice;the kinetics of tumor formation and tumor growth were measured,and tumor growth inhibition rate was calculated;Tumor tissue samples were taken and examined to assess the inhibitory effect of LiCl on tumor growth in the mice. Results LiCl could significantly prevent the proliferation of SMMC-7721 cells and showed concentration dependency. It could also induce apoptosis in SMMC-7721 cells;Marked inhibitory effect of LiCl on transplanted hepatocellular carcinoma H22 was observed in the tumor-bearing mice, the inhibitory rates were 50.0% and 60.7% in the groups treated with middle and high dosage of LiCl. Apoptotic cells and apoptotic bodies were observed in the groups treated by LiCl under the electron microscope. Conclusion LiCl had marked inhibitory effect on the proliferation of SMMC-7721 cells and the tumor growth of hepatocarcinoma-bearing mice which was probably related to induction of apoptosis.

    Objective To observe the changes of the biochemical index and pathology of the rabbit liver acutely poisoned by poisonous mushroom. Methods The rabbits were i.g with toxins from Amanita fuliginea at the dosage of 0.05、0.1, 0.2g/kg respectively, and the matched control with 0.9%NS. The biochemical index of liver were observed in every two days, the rabbits were sacrificed after 120 hours, and the pathologic changes of the liver were examined. Results Liver function of the control is normal. Liver function of test groups were damaged with different degree, degeneration or necrosis happen in liver cell, inflammatory cells are in a vessel or a leaf. Conclusion Aniatoxins can damage to liver function of rabbit, and the injury is more obvious depended on the amount of toxins. For making the proper animal model, acutely poisoned by Amanita fuliginea, should choose to give the toxins at the dosage of 0.1g/kg.

    Objective A mathematical model for hypoxic response of the central nervous is presented Method The model is built on Matlab/Simulink system. It combines the models of heart,systemic and pulmonary circulation systems together. The model is composed of carotid sinus baroreceptor afferent path,synpathetic nerve together with hypoxic response of the central nervous system and reflex effector. Results The simulation experiments showed that hypoxic response of the central nervous system plays an inportant part in cardiovascular system. Conclusion The(feveloped model was an effective means to simulate the kinetics of hypoxia.

    Objective To investigate the effect of propofol on TNF-α、IL-1β and IL-10 of endot-oxic shock rat. Methods Twenty-four male Wistar rats were randomly assigned to three groups: Control group(Group C), Endotoxin group(Group L),and propofol plus Endotoxin group (Group P). Endotoxin with concentration of 10 mg/kg were injected into the rats of group L through femoral vein;The rats in group P had 20 mg/kg/h of propofol injected intravenously and then received 10 mg/kg/h of endotoxin; The rats in group C had the same amount of normal saline injected. The rats were killed 60,120,180 and 240 minutes after the injection. The levels of TNF-α、IL-1β and IL-10 were measured by ELISA. Results In Group L, the concentrations of TNF-α、IL-β and IL-10 rose significantly (P<0.001). In Group P, all the concentrations were significantly lower than that of Group L (P<0.001).Conclusion Propofol can significantly decrease the level of TNF-α、IL-β and IL-10 in endotoxic shock, and then prevent acute lung injury caused by endotoxin.

    Objective Study on the effect of phytase on apparent digestibility of crude protein and utilization of calcium or phosphorus in different ages rhesus monkey. Methods Sixteen healthy young and adult breeding rhesus monkeys were selected, and randomly divided into young or adult of treated and control group. Results The apparent digestibility of crude protein were 88.43% ±1.83% and 81.39%±2.28%,in young of treated and control group respectively and the calcium and phosphorus utilization were 61.70% ± 3.70%,84.01% ±2.84%,and 45.32% ± 9.63%,71.15% ± 4.67%,respectively whereas in adult were 89.28% ±1.63% and 81.65% ± 2.95% ,34.18% ± 10.41%,81.97% ±3.38% and 21.18% ± 6.56%,69.23% ± 9.15%. Not only P<0.05 for the comparison with the control of phosphorus utilization in the adult, but also P<0.01 of calcium or phosphorus utilization in young group and crude protein apparent digestibility of all. Conclusion The phytase has a good value on utilization of young rhesus monkey.

    Objective Discuss the safety of the ‘PiGuShiEr’(a special dispensation) on mice. Methods Feeding with the dispensation of ‘PiGuShiEr’ every other day, the effects on mice were observed including the increment of body weight, food efficiency ratio (FER), viscera index (VI), hemogram and metabolism mark of liver and kidney. Results ‘PiGuShiEr’ have a good FER(P<0.05) and ‘PiGuShiEr’ can reduce the increment of body weight, blood-sugar lever, and TG (P<0.05) without impairing the quality of living, hemogram and the metabolism mark of liver and kidney. Conclusion The ‘PiGuShiEr’ is a feasible and safe regimen for experimental mice, and it might improve the metabolism of liver and kidney to be beneficial of metabolic disease.
