Protective Effect of Propofol on TNF-α、IL-1β and IL-10 in Endotoxin Shock Rats

Abstract: Objective To investigate the effect of propofol on TNF-α、IL-1β and IL-10 of endot-oxic shock rat. Methods Twenty-four male Wistar rats were randomly assigned to three groups: Control group(Group C), Endotoxin group(Group L),and propofol plus Endotoxin group (Group P). Endotoxin with concentration of 10 mg/kg were injected into the rats of group L through femoral vein;The rats in group P had 20 mg/kg/h of propofol injected intravenously and then received 10 mg/kg/h of endotoxin; The rats in group C had the same amount of normal saline injected. The rats were killed 60,120,180 and 240 minutes after the injection. The levels of TNF-α、IL-1β and IL-10 were measured by ELISA. Results In Group L, the concentrations of TNF-α、IL-β and IL-10 rose significantly (P<0.001). In Group P, all the concentrations were significantly lower than that of Group L (P<0.001).Conclusion Propofol can significantly decrease the level of TNF-α、IL-β and IL-10 in endotoxic shock, and then prevent acute lung injury caused by endotoxin.
