Objective To investigate the endogenous circadian expression of the Arylalkylamine N-acetyltransferase(NAT)gene in the rat pineal gland.Methods 36 SD rats under constant darkness(DD)for eight weeks were sampled for the pineal gland once every 4 hours in a circadian day.The total RNA was extracted from the samples and the semi quantitative reverse transcription polymerase chain reaction(RT-PCR)was used to determine the temporal changes in mRNA level of the NAT gene during different circadian times(CT).The data parameters were obtained and analysed by both the cosine function software and the amplitude F test to reveal the circadian rhythm.Results The NAT mRNA level in the pineal gland showed a significantly circadian oscillation(〈i>P

Objective To develop the endometriosis model in the mice, and to investigate the methods of analyzing the ectopic endometrium using the fluorescent imaging. Methods Endometrial fragment obtained from the trans gene tic mice treated by Green Fluorescent Protein (GFP) and injected into the peritoneal cavity of C57BL/6 mice. These recipient mice were raised with water supplemented by E

for three weeks. Then the mice were euthanized,and we observed the intensity of the GFP of the peritoneal cavity under fluorescence microscopy. Results The green fluorescence does exist in the peritoneal cavity of the mice and we have proved that the ectopic endometrium expressed the GFP by the immunohistochemistry. Conclusion The model developed by using the transgeneic mice expressing GFP would be available for exact evaluation of endometriotic lesions, which is prospective.

Objective To establish chronic aristolochic acid (AA) nephropathy in rats. Methods Male SD rats were randomly divided into control and model group. The model group rats were given daily with AA 5 mg/kg intragastically,the control group rats were given equal amount of normal saline. Objective To evaluate the influence of the insulin on the development of mouse preimplantation embryo and try to find the IGF-1 express situation in mouse tissues. Method Two-cell mouse embryos, collected from oviduct flushing,were divided into two groups, one group of embryo were cultured for 36 hours in the KSOM media supplemented with insulin at concentration of 0.25 ug/ml as insulin group (Ins group). Another group of embryo was cultured in the same KSOM media but without insulin as control group. After the embryo cultured to blastocysts in vitro, two groups blastocysts were transplanted to the uterine of induced pseudopregnant ICR mouse .The birth weight of the FI fetal mouse were measured. The IGF-1 mRNA express of two group fetal mouse in their three kinds of tissues (liver、muscle and brain) were detected by half-quantities RT-PCR method. Result The rate of Ins-group development to blastocyst after the cultured 36h is higher than that of control group about 9.7%. The average body weight of FI fetal mouse in Ins-group is higher than that of the control group about 17.8% (
P
<0.01). Half-quantities RT-PCR results also showed that IGF-1 express level in the muscle of FI Ins-group is higher than that of Control group. But no IGF-1 express difference was found in the liver and brain Conclusions The suitable concentration of insulin in the media supplement in vitro can accelerate the embryo development and increase the IGF-1 mRNA express in muscle of the fetal mouse.

Objective To observe the function of anti-inflammation,abirritation and lowering uric acid of effective fractions in Modified “Si miao Powder”. Methods The pain reaction of mouse by chemical stimulation,the ear swelling of mouse induced by xylene and hyperlithuria model were adopted to observe the anti-inflammation,abirritation and lowering uric acid effect of Modified “Si miao Powder” and its effective fractions. Result Modified “Si miao Powder” can fall the uric acid in 3h to the hyperuricemia mouse (

<0.05) and has obvious paregoric effect to the writhing response mouse stimulated by acetic acid (

Murine Norovirus l(MNV-l) is a newly recognized pathogen of mice that causes lethal infection in mice deficient in components of the innate immune response. Moreover, the murine virus 1 is the only norovirus that can replicate in cell culture and in a small animal.
