    Cardiopulmonary Bypass Model in Neonatal Piglet

    The cardiopulmonary bypass models in neonatal piglets were established. Two schemes of protection on immature myocardium were compared in these models with micro-software, MBP-11 multiple channels physiological analytic system. The results showed that the blood cardioplegic solution enriched by amino acids and combined with warm induction/reperfusion were superior to the traditional cold crystal cardioplegic solution.And it is suggested that the cardiopulmonary bypass model in neonatal piglet supported by microcomputer is an effecient means for the research in the protection of immature myocardium.

    To study the mechanism of protective effect of dexamethasone on myocardial reperfu-sion injury,the isolated working heart models were prepared in 30 SD rats,and then divid-ed into 3 groups:normal control group (gruop 1), ischemia and reperfusion control group (group 2) and experimental group (group 3). Cold crystalloid cardioplegic solution was perfused into the aorta of group 2 and 3 before cardiac arrest.Dexamethasone was added to the cardioplegic solution in group 3 at the concentration of 20 μmol/L. The levels of nitric oxide (NO) and creatine phosphokinase (CPK) in the blood of coronary sinus,and of NO synthase (NOS) and malondialdehyde (MDA) in myocardium were measured before arrest and 30 min after coronary reperfusion. The cardiac function was assessed at the same cor-responding times. The results showed that in group 2,as compared with group1,the levels of NO and constitutive NOS (cNOS) were reduced and inducible NOS (iNOS) increased after cardiac reperfusion,in the experimental group,as compared with group 2,the levels of NO and iNOS were reduced, while cNOS not significantly changed,and the recovery of cardiac function was ameliorated,the levels of MDA and CPK were reduced. These indicated that dexamethasone was helpful for myocardial protection. The reduction of iNOS and NO induced by dexamethasone may play an important role in myocardial protection.

    The activities of alkaline phosphatase (AKP) and acid phosphatase (ACP)in the hearts,livers and kidneys of KM mice at different stages of development were measured with enzyme histochemical method at 4,8,12,24 and 36 h after the mice being subjected to heat shock(44℃,30 min). The results showed :① The sensitivities of mice to heat shock at different stages of development were differed. The mice after birth were more sensitive than the fetuses.② Heat shock could induce the expression of AKP in the heart.③ Heat shock induced significant increase in the activities of AKP and ACP in the liver and kidney,and of ACP in the heart. The results suggested that AKP and ACP may play an important role in maintaining the normal physiological function of the cells in the heat shock responses.

    The models of partial thickness burn wound were prepared with SD rats.The mRNA expressions of 4 growth factors in the exudate cells were analysed quantitatively with RT-PCR method during the wound healing. The results showed that the expressions of TGF-α,TGF -β1 and bFGF were observed at 6h after scald,the expression peaks of TGF-α, TGF-β1 and PDGF were observed 3d after scald, the expression peaks of TGF-β1 and bFGF were observed 10d after scald,and the levels of these peaks were significantly higher than those in the control group (P<0.05).The results would be helpful for the further in-vestigation of the mechanism of burn wound healing.

    To investigate the effects of all-trans retinoic acid (ATRA) on the growth of gastric cancer cells in nude mouse xenografts,three gastric cancer cell lines,MGc80-3,BGC-823 and SGC-7901,were treated with 10-6 mol/L ATRA for 20d,and then inoculated subcu-taneoiLsly in the right hind legs of nude mice respectively,while the untreated ones in the left hind legs. Four weeks after the implantation,the nude mice were executed,the xenografts were removed and examined pathologically. The results showed that the incuba-tion periods and/or the double times of the xenografts of the treated group were prolonged,and their growth was inhibited significantly with inhibitory rates of 51.4% (MGc80-3) ,66.0% (BGC-823) and 88.6% (SGC-7901) respectively,These indicated that ATRA treating in vitro could inhibit the growth of the gastric cancer cells in nude mouse xenografts,and the degrees of inhibition were negatively correlated with the malig-nancy of the cancer cell lines.

    The method of PCR for detection of infectious ectromelia (Ect) virus was established by a set of consensus sequence primers. The results showed that the PCR amplified product was 846 bp and verified as Ect virus by restriction endonuclease analysis,and that the method could detect Ect virus gene even at the level of 0.lpg.
