    Selection and Establishment Highly Metastatic Model of Human Melanoma in SCID Mice

    We have successfully established a highly metastatic model of human melanoma in scid mice (SCI-375). The selection of metastasis was obtained by using successively subcutaneous implantation of lungs metastatic tissue in scid mice. The metastatic rates were 100%and 80% respectively in lungs and lymph node from 6-21 passages. The lung metastatic tumors were similar to the original human melanoma in histopathological features and chromosomal karyotype. This metastasis model may be used as a tool for the research on the metastatic mechanism and treatment of the human tumor.

    To study the inflammatory reaction and lung injury in pneumocystis carinii pneumonia (PCP),PCP was induced in Sprague-Dawley rats by the administration of cortisone acetate as immunosuppressant for 8-12 weeks.The Rats were divided into four groups,Group Ⅰ: normal control (n= 6),consisted of healthy rats without the steroid treatment ; group Ⅱ:negative control (n= 6),rats free from lung infection; group Ⅲ : bacterial pneumonia (n= 11); group Ⅳ (n= 14) : PCP. Cells in the bron-choalveolar lavage fluid (BALF) were counted and differentiated,as well as the amounts of total protein (TP), albumin (ALB) and the activities of alkaline phosphatase (ALP), lactate dehydrogenase (LDH)and type Ⅳ collagenase (matrix metallo-proteinases, MMP-2,MMP-9) were detected. And compared the differences in inflammatory reaction and lung injury between PCP and bacterial pneumonia . The total cell count in the BALF of PCP was significantly increased (P<0.01),but was lower than that in the bacterial pneumonia group. ALB in the BALF of PCP (622.5±484.0μg/ml) was increased as compared with the negative controls (35. 8±16.2 yg/ml,P<<0.01).The activities of ALP and LDH in PCP (217.0±81.4u/L,20.8±8.2 u/dl respectively) were also higher than those in the negative control (P<0. 01). The activities of Type IV collagenase were increased in PCP (P<0.001). The inflammatory response in PCP differed from that found in bacterial pneumonia. The permeability of the alveolar capillary membrane and the lung injury were significantly increased in PCP.
    Cryopreservation of MMTV-Wnt-1 Transgenic Mice Embryo by Using High Concentration Ethylene Glycol

    The high concentration ethylene glycol (EG-8,8 mol/L) and galactose(1.7mol/L) were used as a cryoprotectant and thawing diluents for embryo cryopreservation by vitrification in MMTV-Wnt-1 transgenic mice. The best results of cryopreservation were obtained if those embryos kept balance in EG-2 (2mol/L) for 4~6 minute before freeze and thawing 3~5 minute in galactose. The percentage of embryos cryopreserved and retrieve is 88. 61% (109/123); there is 88. 99%(97/109)of these embryos develop and 81. 65%(89/109)developing to hatch blastocysts. After these development embryos were transfer to pseudocyesis mouse, 38. 36%(69/181) developed to live young, and a half of these is positive mouse on the PCR test. It follows that high concentration ethylene glycol and galactose as a cryoprotectant for cryopreseveration by vitrification is effective.
    Ultrasound-guided Establishing of VX2 Liver Cancer Model in Rabbit

    To establish VX2 liver cancer model in rabbit,the VX2 tissue was cut into strip shape about 1 mm× mm×3 mm, and were inoculated into the liver of 60 New Zealand rabbits guided under ultrasound. The successful rate of the animal model were observed 2 weeks later. Spherical or sphere-like hypoecho lesion could be found in the model. No echo of envelope could be found at the inoculated site. Heteroecho was observed when the tumor has coliquation and necrosis. The cross section of the tumor was hoariness and stiff, with fish meat-like appearance. Natural life of 9 model animals were 36~52(43.3 ±5.7)days. The results suggested that the ultrasound-guided percutaneous puncture-inoculation method for establishing rabbit VX2 liver tumor model is simple and effective.
    Allergic Rhinitis Model Induced by Ragweed Pollen Allergen in Guinea Pig

    To establish an allergic rhinitis model induced by ragweed pollen allergen, guinea pigs(n=42) were divided into a sensitized group(n=32)and an unsensitized control group(n=10). Sensitized groups were immunized intraperitoneally and intracuta-neously with ragweed pollen allergen for 4 weeks, followed by intranasal administration. Unsensitized control group immunized with 0.9% NaCl instead of ragweed pollen allergen. As a results,symptoms of allergic rhinitis,such as sneezing,itching and rhinor-rhea were observed in 28 guinea pigs of sensitized groups. There were significant increases in the number of eosinophils and Mast cell in the epithelium after 30 min in the sensitized group received intranasal ragweed pollen allergen. So, An animal model of allergic rhinitis induced by ragweed pollen allergen was established successfully. It provides a tool for studying and treating allergic rhinitis’especially induced by ragweed pollen allergen.

    We constructed an experimental rat carotid artery stenosis model induced by FeCl3. The smooth muscle cells migrate from the median to form a neointimal after the endothelial cells are injured by FeCl3. The FeCl<sub>3</sub> injury model has the same pathology as balloon injury model, and easier to operating, and also the degree of injury can be controlled quantotatively. It will be useful in screening new drugs and pharmacodynamic study.

    Recent years,in spite of all its limitations and high expenses,the nude mouse-human tumor model has become more frequently applied to the evaluation of new compounds and pre-clinical research. We reviewed briefly the pre-clinical criteria of NCI and EORTC investigation in this paper,and hope to consult for researchers in our country.
