    Cloning and Sequencing of the nuc Gene of Staphylococcus aureus

    According to the sequence of the nuc gene, which encodes the thermostable nuclease (TNase) of Staphylococcus aureusS. aureus), a pair of oligonucleotide primers which are special to the sequence of nuc gene were designed and synthesized. Using the genome DNA of the clinically isolated strain of S. aureus SA. m0109 as template, the fragment flanking nuc gene was amplified by Polymerase Chain Reaction (PCR). The expected size fragment was obtained and was cloned into the plasmid pGEM-T. The recombinant plasmid of pGEM-NUC was identified by restriction enzyme analysis, PCR and sequencing, which proved completely its validity.
    Influence of Epidermal Growth Factor on Signal Conduct Mechanism of the Tongue-coating Formation in mice

    To research the signal mechanism between epidermal growth factor (EGF) and tongue-coating formation,the mice tongue mucosa cell growth with EGF-R expression was observed after droping EGF to mice mouth cavity.HE、EGF-R immuno-histochemistry (SABC) and by data handles system (CMIAS-98A) to proceed the image analysis were used. The results showed that EGF promotes the thickening of tongue mucosa in experiment mice,and the base layer cells and the EGF-R expression of tongue mucosa were increased.The EGF might influence the tongue-coating formation by the signal conduct mechanism of EGF-R.

    The scanning electron microscope,transmission electron microscope observations revealed that the lingual ultrastructure of miniatue pig is very similar with that of hunam being. According to anatomic and histological results,miniatue pig is completely fit to be selected as an ideal pathological animal model.To explore the method of establishing greasy coating model,different ways were performed and finally the animal model is successfully established by giving a mixture of 50% spirits and edible alkali saturated solution intragastrically every other day.

    To study the histopathological changes in spontaneous kidney lesions of breeding maca-ca mulatta,kidney samples from the monkey at the age of 2~20 years were investigated by routine histological technique.The results showed that various spontaneous lesions including glomerulus’s,renal tubules and interrenal tissue etc, totaling more than 14 kinds in different degrees ,were found in 79. 4% of the monkeys. The degree and the frequency of the lesions were increased with age. The results suggested that the spontaneous lesions of kidney were often ob-served in macaca mulatta and its interference effect on medicine research should be considered se-riously.

    Two kinds of specific and sensitive methods for detecting Toxoplasma gondii was estab-lished by means of NT-PCR and One-Tube Hemi-Nested PCR. The DNAs were extracted from Toxooplasma gondii、Giardia lamblia and also blood、brain、liver、spleen、lung、kidney tissue of ICR mice infected with Toxoplasma gondii by routine methods. Two DNA fragments of the expected size (522 bp and 362 bp) was ampliflied from Toxoplasma gondii、blood、brain、liver、spleen、lung and kidney tissue by using NT-PCR and Hemi-Nested PCR rspectivelly, but no band was observed in Giardia lamblia. The sensitivity of the reaction was determined with different concentrations of genomic Toxoplasma gondii DNA,the DNA even less then lpg could be detected by us-ing single-step PCR.The NT-PCR or One-Tube Hemi-Nested PCR was more sensitive than sin-gle-step PCR.

    To establish the model of restenosis,the metal stent was implanted into the canine femoral artery through the carotid artery.Twelve weeks after the stent implanted,the lumen of canine femoral artery was reduced significantly as a result of smooth muscle cell proliferation and neointima formation. This model is reliable, easier to operate,and resembling the same pathologi-cal changes of clinical restenosis. It is an ideal model for restenosis research.

    Establishment of Rabbit iliac artery restenosis model after percutaneous transluminal coronary angioplasty (PTCA)is the base for research the mechanism of restenosis. According to the character of coronary artery of Chinese people, we established an animal model by using 2.5-mm balloon catheter to denude endothelium in rabbits iliac artery. Twenty eight days after operation ,the tissue sections were stained with HE (hemalaune hematoxylin and eosin) and van Gieson for morphologic examinations. Smooth muscle cells (SMC) were indicated immunohis-tologically with an antibody against a-acting .The indexes of stenosis and proliferation were determined by image analysis system. The result showed that the model can simulate the pathologic procedure of restenosis after PTCA and confirmed that the proliferation of SMC is the mainly cause of restenosis.

    Nine random primers were used to amplify genomic DNA with randomly amplified polymorphic DNA (RAPD) methods in 3 inbred strains of rats. The result showed that the amplified band patterns of every individuals were same in one strain,the 3 strains of rats were homogeneous at these RAPD markers, however, the amplified band patterns showed different among three strains. The similarities between Wistar arid WKY, Wistar and F344,WKY and F344 were 87. 9%、98.5% and 89.2% respectively. The result indicated that the RAPD technique can be used as a method to monitor genetic contamination in inbred rats and to distinguish different strains of rats.

    A Study on Myocardial Injury from Defibrillation of Monophasic and Biphasic Shocks

    During the defibrillation of monophasic and bipliasic in 30 pigs, three methods for the e-valuation of myocardial injury were used:pathological examination, defibrillation electrocardiogram recording,and creatine phosphokinase (CPK) and CPK-MB detection before and after defibrillation. Through visual observation and microscope examination, monophasic and biphasic shocks may cause endocardial edema, thrombosis, localized fragmentation of myocardium and retrograde changes. But the result in biphasic shock was better than monophasic’s. Soft myocardium and endocardium with thrombi was detected in 4 pigs (9-12 shocks,20-50J). Fragmentation of myocardium was observed in the surface of 5 pigs’ defibrillation (5 shocks, 360J). In the biphasic and monophasic defibrillation, retrograde changes were detected in 2 pigs.ST-T seg-ment changed in electrocardiogram in 4 pigs and blockade appeared between ventricle and auricle in 2 pigs after monophasic shock. In biphasic shock,2 pigs and 1 pig obtained respectively. Be-cause CPK and CPK-MB varied in large range,the dates were not evaluated. This results suggest that the biphasic shock cause lower myocardial.injury than monophasic shockdo,with heat and electricity as possible causative factors. When the total energy is same,the defibrillation with high energy and little times can cause bigger myocardial injury than the defibrillation with low energy and more times;the injury may be healed.

    It is necessary to prepare suitable animal model for the study of hypoxic-ischemia reperfusion brain damage. Bilateral common carotid arteries in 7-day-old SD rats were clipped by mini vascular clips for 1 hour,at the same time, placed rats into the hypoxic device for 2 hours to prepare the neonatal model with hypoxic-ischemia feperfusion brain damage. The model was proved to be successful according to pathological exams,the characteristic expression of cyclooxygenase-2 gene and the fine recanalization of the common carotid arteries.
