    Preliminary Study of Pathogenicity of Avian Influenza H5N1 Virus to Gerbil

    Objective To study the pathogenicity of avian influenza H5N1 virus to Gerbil. Method Anesthetized gerbil were inoculated intranasally with avian influenza virus isolates at a biosafety level 3 laboratory. Throughout the 14-day experiment, clinical signs were observed; body weight and body temperature were recorded; the blood was collected to analyze AIV antibodies;tissue samples from the lungs, livers, kidneys and so on were processed for virus isolation and histopathology. Result The H5N1 AIV caused severe diseases in gerbils mainly between day 2 p.i and day 6 p.i,characterized by ruffled fur, inappetence, hunched-back posture, labored breathing, hypothermia, weight loss and 44% mortality. Antibodies were detected from 8 days p.i. The histopathologic examinations showed that variable degrees of lesions including congestion and hemorrhage were observed in the lungs, livers, kidneys and so on.. The lesions of the lungs showed interstitial pneumonia, dropsy, configuration breakage. Conclusion Avian influenza (H5N1) virus is pathogenic to Gerbil.
    Nuclear Factor-κB Plays an Essential Role in Early Phase of Myocardial Ischemia/Reperfusion Injury

    Objective To elucidate the role of nuclear factor kappa B (NF-кB) in early phase of myocardial ischemical reperfusion injury. Methods Eighteen goats were randomly divided into three groups:extracorporeal circulation group(CPB group), ischemical reperfusion group (IR group) and ischemical reperfiision plus pyorrole dithitocarbamate group (PDTC, a special inhibitor of NF-kB activation). Goat hearts were subjected to 60 minutes of ischemia, followed by 90 minutes of reperfusion(the inhibitor team goats were administered with PDTC before cardiac arrest), then hemodynamics of data and cardiac function was measured. Cardiomyocyte apoptosis index was determined by Tdt-mediated dUTP nick end labeling and NF-kB binding activity in the nucleus was measured by electrophoretic mobility shift assay (EMSA) analysis. Results Ischemia/reperfusion could lead to typical histological change of myocardial ischemic injury, while NF-kB was obviously activated in the ischemic injury. The levels of NF-kB binding activity in the myocardium increased signifficently in contrast to isolated extracorporeal circulation group (P<0.05). However, 60 minutes or 90 minutes after reperfusion, the levels of NF-kB binding activity in ischemic reperfusion with pyorrole dithitocarbamate group and the histological change of myocardial ischemic injury reduced signiflcently. Apoptosis index of the ischemic myocardium from IR and PDTC groups was 11.2%±0.4% and 6.35%±0.2% respectively (P<0.05). Conclusions Nuclear factor-KB plays an essential role in the early phase of myocardial ischemia/reperfusion injury, PDTC reduces myocardial ischemia/reperfusion injury by inhibiting activation of transcription factor NF-kB.

    Objective According to the free radical theory for aging, to establish the simple and easy method for brain-aging model, providing theory for brain-aging mechanism and clinic prevention and therapy. Methods The acute brain-aging model was established by 60Co radiation, the second-acute one was established by injection of D-gal, and 5-monthly SD rats as the young control group. The rats, learning and memory ability was determined by Y type labyrinth, the rats’ antioxidant and lipid peroxidative injury were measured by SOD and MDA in blood. The neuron’s lose, injury and degeneration were observed with microscope. Results We found the descend of brain aging rat’s learning and memory ability with Y type labyrinth;The blood’s SOD in brain-aging model was significantly lower than that of the control, the MDA content in brain-aging model was markedly higher than that of the control; The neuron’s lose, injury and degeneration were observed by microscope in different brain region of the rat model, especially in cortex and hippocampus mostly. Conclusions The acute brain-aging rat model can be established simply and easily by radiation.
    A Model of Heart Failure Induced by Volume Plus Pressure Overload in Rabbits

    Objective To establish animal model of heart failure induced by volume plus pressure overload in rabbits. Methods Twenty-two rabbits were divided into two groups: 11 rabbits with heart failure and 11 sham-operated animals. We first obtained a volume overload by creation of an aortic insufficiency, which was followed 14 days later by pressure overload induced by constricting the abdominal aorta. The homodynamic parameters and cardiac structure and function were detected via catheterization and echocardiography respectively 6 weeks later. Results (1)Aortic systolic pressure, pulse pressure, left ventricular end diastolic pressure of heart failure rabbits were significantly increased compared with sham-operated rabbits (P<0.01). (2)The left atria diameter, left inter-ventricular septum, left ventricular end diastolic and systolic diameter were also increased markedly in heart failure rabbits, but their ejection fraction and left ventricular shorten fraction were decreased (P<0.01). (3)In heart failure rabbits, the ratio of left ventricular mass and pulmonary mass to body weight were higher than that in sham-operated rabbits (P<0.01). Conclusion Animal model of heart failure were established by volume plus pressure overload in rabbits successfully, with short time, better repetition and higher rate of success, according with the development of human heart failure.

    Objective To establish the mice model of multidurg resistance of tumorous expression in ascites with SI80 cell. Methods The mice with SI80 cell in ascites were treated with the method of PFC of repeated-combined-chemotherapy in less than the therapy dose for 4 weeks. The levels of P170 and LRP and TOPOⅡ were detected by Flow Cytometry Analysis. Results The cellular multidurg rdsistance of tumorous expression (P170 and LRP) and the activity of TOPOⅡ of mice were improved obviously and the expression was stable in the cell line.

    Objective To satisfy the need of clinic diagnosis, a rapid samples preparation method of rat tissues for TEM observation by microwave process is investigated. Methods The samples of from viscera were divided into three groups numbered Ml, M2 and M3 according to the difference in processing time and microwave power. Results The ultrastructures of the cells can be preserved intactly as original state and a clear ultrastructure and boundary can be observed in all groups. For M2 group, no evident ultrastructural difference can be seen between processed samples and normal samples. But for other groups, slight swelling of mitochondrion from liver and heart in M1 group and slight scathe from organoid of liver, kidney and heart in group M3 were found. Conclusion The preparation time of electron microscope samples can be greatly shorteneed by microwave process on the premise of intactly preserving ultrastructures of viscera cells, therefore shorten the diagnosis time in clinic.

    Objective To establish the rat model of Multiple Organ Dysfunction Syndrome (MODS) caused by trauma. Method Sixty SD rats were divided randomly to MODS model group and control group. The rats of the MODS model group were scalded at 30% TBSA Ⅲ degree and injected with large amount of LPS 6 hour later. The mortality in 7 days, the blood ALT,CK,BUN, fasting glucose and TNFa, NO, IL-6 of MODS rats were observed or determined. Results The mortality in 7 days of the MODS rats was 53.33%. The blood ALT, CK, BUN, fasting glucose and blood TNFa, NO, IL-6 of the MODS rats were higher than that of the control. Conclusion Serious scalded rats with large amount of LPS injected can represents a characters of MODS.

Further more, large numbers of embryos can be produced year round and the optically transparent embryos fertilize and develop externally. All these advances have made this small aquarium fish a new “pet” of toxicologists. Zebrafish has been widely utilized in embryonic and developmental toxicology, environmental toxicology, pathological toxicology and pharmaceutical toxicology research.
