    Establishment and Preliminary Application of Nested Polymerase Chain Reaction for Bacillus piliformis

    On the basis of the 16S ribosomal DNA(rDNA) sequences, the DNA extract-ed from Bacillus piliformis,Escherichia coli,HemoLytic streptococcus,Pasteuralla pneumotroopica were amplified by PCR using 2 pairs of PCR specific primers. With out-er-primers,a DNA fragment of the expected size (625bp) was amplified from Bacillus piliformis and E.coli, but no band was observed in Hemolytic streptococcus and Pasteu-ralla pyieumotroopica. Whereas a 196bp fragment was obtained only from Bacillus pili-formis with inner-primers. Thirty liver samples from immunosuppressive rats were de-tected for Bacillus piliformis by using nested PCR, the positive rate is 30% (9/30),but no serum sample is positive monitored by IFA. In conclusion, the 16S rDNA-based PCR established is sensitive and specific in detection of Bacillus piliformis in infected ani-mals.

    To explore and develop a practical way for hand rearing tree shrews, which is one kind of very useful animal in medical research. 5 pairs of adult tree shrews were housed in breeding cages, fed on the food mixed with purified standard diet, cheese,fruits and vegetables. Newborn tree shrews were moved out from their mothers' cages soon after birth and kept in an incubator for about 3 weeks. The infant tree shrews were fed on a milk formula for the first 3 weeks, then they were moved out from the incubator and fed on a transitional diet for 10 days when they got ready for adult food. There were 31 pups in 11 litters were born in 7 months by 5 pairs of adults, in which 27 pups were hand-reared from birth and 25 of them survived to 7 weeks of age when they were used in an experiment. The average period between the day of pairing and the day of parturition was 52 ± 8. 9 days. The average litter size was 2. 8±0. 4. The results showed that the survival rate of hand-rearing the newborns in this experiment was 92.6%, which is higher than those reported in related publications. The success in hand rearing tree shrews would make it possible to meet the increasing demand of tree shrew in medical research, especially in the field related to liver cancer and hepatitis.
    Determination of Some Haemal Physiological Parameters in Ba-ma Mini-Pig

    Twenty-four haemal physiological parameters in Ba-ma mini-pigs (♂20,♀30)from 4 to 12 months of ages were determined and analyzed. The results showed that no significant difference were observed between sex in haemal physiological parameters in Ba-ma mini-pigs. Whereas most of the detected items were different among the different months of ages (P<0. 05 orP<0.01). Moreover, the values of almost half of the items showed increasing or decreasing with the increase of months of ages. The majority items of Ba-ma min-ipigs were similar to those of common pigs, and 14 haemal physio-logical parameters of Ba-ma mini-pigs were similar to those of human beings,for cxample, Neutrophilic granulocyte, Lymphoyte, Monocyte, Eosinophil, Basophilic cell, Hemohlobin concentration etc. It is concluded that there are some sinilarities in physiological charactcrs betwcen Ba-ma mini-pigs and human beings.

    Alzheimer's disease(AD)is a progressive meurodegenerative disease. It is dnown that AD is associated with some neuotransmitters disorder. We examine the monoamine transmitter in neuons of forebrain by HPLC-ECD method. The content of the 5-HIAA in APP amyloid precursor prorein 751 trangenic micc show significant difference compared with that of the control group. The content of the DA and 5-HT in APP695 transgenic mice increased 38.9% and 45.8% respectively compared with control group. It suggested that the DA and 5-HT system may be affected respectively by Aβ

    To explore the feasibility of replacing nephrectomy of the recipient native kid-ney with extracorporeal ligation (EL) of the recipient native renal vessels for kidney transplantation in rats, The EL of the left and right renal vessels were performed re-spectively at an interval of 20 days in 10 Sprague-Dawley rats of nontransplant group. The EL of the right native renal vessels were performed in 10 kidney transplant Lewis rats with resected left native kidney 3 to 5 days after kidney transplantation. The results showed that the nontransplant rats survived well after EL of the left renal vessels and died within 3 to 4 days after EL of the right renal vessels. The transplant rats survived well after EL of the right native renal vessels. The pathological examination of the kid-neys with EL showed extensive necrosis and that of the transplanted kidneys showed normal structure. It can be concluded that the EL technique of the rat renal vessel is simple, reliable and little traumatic to rats, thus it may be used to replace the nephrec-tomy of the recipient native kidney for kidney transplantation in rats.
    The Effects of Mifepristone on the Cerebral Tissue of Term Pregnant Rats

    The effccts of mifepristone on the cerebral ultrastructure of term pregnant rats were studied. Ten pregnant rats were divided randomly into an experimental group (n = 5) and a control group (n = 5) . At the 20th day of gestation, the experimental rats were given sesame oil 1 ml and mifespristone 11.5±1mg, while the control rats were-given only sesame oil 1 ml. After labour, the cortical tissues of the right parietal cere-brum were obtained and prepared for electron microscopic observation and analysed by stereology. Results :compared with the control group, the experimental group showed the following morphological changes: (1) Neurons swelled and membranous cytoplos-miz organoids decreased ; (2) Neuropil was partially damaged and major dense lines of myelin sheath were separated; (3) Synaptic connections decreased; (4) Interstitium was swollen ; (5) The numerical density of mitochondria in the experiment group was de-crease,but the volume density showed no significant change. Conclusion : Mifespristone could cause ischemia-hypoxia changes in cerebrum of pregnant rats.

    In five consecutive years,5319 serum samples were collected from different species of conventional, clean and specific pathogen free (SPF) laboratory animals in twenties facilities for the detection of viral antibodies. The results showed that the posi-tive rate of the antibody to Mousepox virus(MPV) was 3.2%(30/927),to Lymphocytic Choriomengitis virus (LCMV) was 0.3% (3/927) and 5.1% (21/914) in conventional mice and guinea pigs colonies, respectively. Antibody to Mouse Hepatitis virus(MHV) was 2.8% (26/914),and to Sendai virus was 2.0% (18/914) , 2.8%(4/141) and 33.3% (4/12) in clean mice,rats and guinea pigs colonies,respectively. Antibody to Reovirus type III(Reo-3) was found in SPF mice colonies kept in isolator with a positive rate of 1.6%(375/381).

    Glycohemoglobin HbAlc of diabetes rats was rapidly determined by capillary electrophoresis to investigate the change of HbAlc of diabetes rats treated with drugs. The results showed that capillary electrophoresis could rapidly and effectively separate glycohemoglobin from non-glycohemoglobin. HbAlc of diabetes group without treatment was significantly higher than those of control,aminoguanidine,tetram-ethylpyrajine and united treatment groups(P<0.01). There were no significant differ-ence among the treatment groups(P>0.05). It suggested that determination of HbAlc by capillary electrophoresis could be used as an indicator of therapeutic efficacy of dia-betes rats.

    Spontaneous neoplastic lesions are often observed in aged experimental ani-mals. The case reported here was a spindle cell sarcoma in mouse. The original site was uterus with distant lymph nodes metaslases. The neoplasma was composed of interlac-ing bundles of elongated spindle cells with palisading of nuclei. Some areas show degen-eration and necrosis. It was not very typical in ordinary appearance of leiomyosarcoma, but a special type of smooth muscle tumours.

    The researches on animal models infected by varicella-zoster virus (VZV) in the latest twenty years were retrospected. The authors summarized the methods used both in the establishment of the models on non-human primates,guinea pig, rat and mouse and the methods for the detection of the virus,and outlined the applications of the models. To compare, summarize and analyze these documents is very beneficial to studying the mechanism of VZV infection and developing vaccine and anti-virus medicines.
