The Expression of Enhanced Green Fluorescent Protein Gene in Chicken Blastodermal Cells

Abstract: To search the appropriate experimental condition for using green fluorescent protein (GFP) as a reporter in transgenic chicken, Freshly laid unincubated eggs from White Leghorn Chicken were used. Only embryos at stage X were used to separate the blastodermal cells.After incubated over night ,the blastodermal cells were transfected with pLEGFP-Cl by lipofectame. Fluorescence could be detected in intact viable cells under different sets of conditions, and the expressed fluoresccnce was stable. The trans-fcction rale of pLEGFF-Cl expressed was different in three cells lines examined. The GFP can be visualized continuously and directly for gene expression in living cells.
