Analysis of the Porcine mtDNA D-loop PCR RFLP

Abstract: To analyze the polymorphism of the mtDNA D-loop in pigs,a pairs of primers were designed to amplify mtDNA D-loop of Xishuangbanna Small-ear pig, Guizhou miniature Xiang pig, Guangxi Bama miniature pig,Rongchang pig,Lang Yorkshire and Landrace. The amplification products were digested by 23 restriction enzymes and detected by 2% agarose gel. Clear amplification band were obtained,and the fragment length was the same as that of being reported before. No polymorphism was found among or in stocks. The results suggested that there is no polymorphism in porcine mtDNA D-loop RFLP,therefore it is not suitable as a method for the differentiation of the breed or stock in swine.
