Establishment of Gene Transfer Model of Vein Autograft in Rabbit

Abstract: The purpose of this study is to develop a new way to transfer extrinsic gene to vein graft. The models of carotid-jugular bypass were prepared in rabbits. The human wild-type p53 gene adeno-associated virus plasmid vector was transfected into vein graft mediated by the cationic liposome DOSPER by using intraluminal pressure perfusion method. The gene expression was determined by polymerase chain reaction technique and immunohistochemistry. It was verified that the extrinsic p53 gene was transduced into vein graft successfully and expressed mRNA and p53 protein. The results suggested that intraluminal pressure perfusion method is one of the efficient means for vein autograft gene transfer system.
