Establishment of Two Human Osteosarcoma Cell Lines and Their Biological Characteristics

Abstract: In order to study the characteristics of two cell lines (S1,S2) derived from human osteosarcoma cell LTH - OS(S) and established by clone technique and transplantation. Cellgrowth in soft agar,chromosome analysis, histological observation, and tumor formation rate in nude mice were used to characterize the cells. Electrophoresis mobility was 1.00 and 0.70,the doubling time of population was 22.4 h and 24.0 h,the clone formation rate in soft agar was 33.5% and 23.1% for S1 and S2,respectively. Extensive lung tumor was observed in nude mice after inoculation of S1 cells through tail vein. The results indicated that the cell lines isolated may be very useful material for the study of characteristics of metastatic cancer cells.
