Detection of Pasteurella Multocida by ELISA Inhibitory Test in Laboratory Animals

Abstract: A rapid sensitive ELISA inhibitory test was designed to detect Pasteurella multocida infection in laboratory animals. In the reactive system, the soluble material extracted from P. multocida(P.m)strain P-1059 X-73 or A-898 was used as coating antigen, rabbits were immunized with these three strains of whole bacteri resapecti-vely. The purified rabbits IgG of anti-P.m was labeled with HRP by sodium periodate method. The HRP labeled rabbit anti-P. m IgG was mixed with animal serum samples to operate ELISA inhibitory test. The conventional indirect ELISA was performed to confirm the results. It was found that infective rate of P.m strain P-1059 in rabbits was as high as 30%; while in mice and hamsters, no positive samples were detected. The results indicated that this is an easy, rapid and reliable method for the detection of P. m infection in laboratory animals.
