    Liver Metastaic human Gastric Carcinoma Established in Nude Mice Using Orthotopic Organ Selection of Metastasis

    Objective To establish a highly liver metastatic model of human gastric carcinoma in nude mice by using orthotopic organ selection of metastasis. Methods Histologically intact MKN-45 human gastric carcinoma tissues were orthotopicly implanted into the gastric mucosa layers in nude mice,repeated screening was conducted according to the method of gastric orthotopic implantation→liver metastasis→subcutaneously inoculation→gastric orthotopic implantation. Tumorgenicity, invasion, me-tastasis and morphological characteristics of the implanted tumors were studied by light microscopy and electron microscopy. Results Highly liver metastatic models of human gastric carcinoma named MKN-45sci were obtained after organ screen. They had been passaged in vivo for 5 generations. 100% tumor take rates were observed, the metastases of liver and lymph nodes were noted in all of the animals. They were also exhibited other features: metastasis of lung、spleen、diaphragm and bloody ascites. The primary and metastatic tumors were similar to the original human carcinoma in histopathological and electron microscopy features. Conclusions The orthotopic implant models of human gastric carcinoma displayed features of human clinical gastric carcinoma in animals. They provided a useful tool for the study of metastatic mechanism and treatment of human gastric carcinoma.

    Objective To investigate inhibition effect of KiSS-1 gene on the trophoblast cell in order to explore the role of high expression of KiSS-1 gene in pregnancy induced hypertension syndrome. Methods Thirty pregnant Wistar rats were divided into three groups randomly with 10 rats in each group. Group A (pcDNA3-KiSS-1 treated group) was transferred recombinant eukaryotic expression vector pcDNA3-KiSS-l encapsulated with lipofectamine by intraperitoneal injection on day 0.5, day 7.5 and day 14.5 of gestation respectively. Group B (pcDNA3 treated group) was transferred pcDNA3 encapsulated with lipofectamine by intraperitoneal injection at the same day. Group C (control group) was injected 0.9% sodium chloride solution at the same day. Blood pressure and urinary protein were assessed serially. All rats were sacrificed on day 21.5 of gestation. Amount of pup, pup weight and weight of placenta were measured. Expression of KiSS-lmRNA and MMP-9niRNA in placenta were determined by reverse transcription- polymerase chain reaction (RT-PCR).Placenta, uterus attached to placenta, kidney, heart and liver were examined histologically. Results(1) Compared with Group B and Group C, Blood pressure in Group A began rise in day 15.5 of gestation (P<0.05), gradually increased to peak levels until delivery. (2)Group A show increased urinary protein in day 14.5 of gestation and persisted to delivery. Daily urinary protein in Group was significantly higher than that in Group B and C (P<0.01) (3) Pup weight in Group A was significantly lighter than that in Group B and C (P<0.05), and resorbed pups were observed in Group A.(4) Expression of KiSS-lmRNA was positive in 7 of 10 placenta in Group A, all in Group B and C was negative. Expression of MMP-9mRNA in placenta was significantly decreased in Group A compared with that of Group B and C (P<0.05).(5)Histological exami-nation show proliferation of the glomerular tufts and thickening of media of the kidney arteriolar wall in Group A compared with Group B and C. There was no significant difference in placenta, uterus attachment to placenta, heart, liver among three group. Conclusion The pcDNA3-KiSS-l vector encapsulated by lipofectamine was transfected and integrated into pregnancy Wistar rats. Expression of KiSS-lmRNA was positive in 7 of 10 pcDNA3-KiSS-1-treated rats while negative in pcDNA3-treated and control. pcDNA3-KiSS-1-treated rats exhibited the feature of preeclampsia including hypertension and proteinuria. It suggests that over expression of KiSS-1 cause deficiency of invasion of trophoblast and insufficient of remodeling of spiral arteries in placenta. It may play main role in shallow implantation of placenta in pregnancy induced hypertension syndrome.

    Objective To explore the sequential pathological changes during the process of establishing high fat diet fatty liver model in rats. Methods Sixty male Wistar rats (180-220 g) were divided randomly into six groups:normal control Groups (1,2,3)fed with common feed and model groups(4,5,6) fed with high fat feed (93.8% foundation feed,1% cholesterol,5% lard,0.2% bilesalt )for 4,8,12 weeks. Results The liver index of the model group 4 is significantly higher than those of group 5,6 and normal control groups;but the liver index of the model group 5,6 is significantly lower than which of the control groups. The histological observation shows that the form of lipid droplet of the liver cell is mainly grain type, small bubble, large bubble in early, medium and advanced stage with the area of the vacuole significantly increasing one by one.The ultrastructural observation on the liver of the model groups' rats shows that the quantity of RER and SER is significantly different in each stage. Conclusion The lipid droplet form ,area of vacuole,quantity of RER and SER should be the evaluating indicator in the estimation of high fat diet fatty liver model,but the liver index and the variety of chondriosome lacks to be.
    Preliminary Study on Effectiveness of RU486 in the Treatment of OHSS Model Rat

    Objective To observe the effectiveness of RU486 in the treatment of OHSS model rat. Methods A total of 30 immature female rats aged 22 days were injected s.c. daily with 10 IU pregnant mare serum gonadotropin (PMSG) for 4 consecutive days from the 22 nd to 25 th day of life. On the 26 th day of life,100 IU human chorionic gonadotropin (HCG) were injected s.c. to produce OHSS model. On the 27 th day of life, 30 rats were randomly divided into five groups. Each group contained 6 rats. The rats in group Ⅰ as control group were not given Ru486 treatment; in group Ⅱ to group Ⅴ were given s.c. with the dosage of RU486 1 mg/kg, 5 mg/kg, 10 mg/kg,20 mg/kg respectively. Fourty-eight hours after RU486 treatment, the ovarian weight, peritoneal capillary permeability and ascites score of all rats were determined. Results The ovarian weight in group Ⅲ and Ⅳ was 0.321±0.027 g, 0.302±0.035 g, respectively, which were significantly lower than that of control group (0.352±0.022 g)(P<0.05,P<0.05 respectively), peritoneal EB content in group Ⅲ and group Ⅳ was 1.18土0.66 μg and 1.17±0.47 μg, respectively, which were significantly lower than that of control group (2.33土1.06 μg) (P<0.001,P<0.001 respectively),and the ascites score in group Ⅲ and group Ⅳ was 2.08±0.38, 1.67±0.48,which were significantly lower than that of control group (2.67±0.41) (P<0.05,P<0.01 respectively).However, there was no significant difference to be found between group Ⅱ and Ⅴ and control group. Conclusion Optimal dosage of RU486 treatment could improve the condition of OHSS in the rat model.

    Objective To explore a new method rearing diamondback moth, Plutella xylostella (L.).Methods Put brassica oleracea seeding into rearing cage directly, let diamondback moth produce eggs on the seeding, and a new method without larva shift was set up. Results The egg hatch percentage is 75.8% 77.4% and the pupa emergence is 79.9% 89.0%. Conclusion This technology has a characteristic of good practicability and is easy to operate for mass rearing.

    Objective To establish a useful experimental system for evaluating the effect of drugs on allergic rhinitis. Method An animal disease model of allergic rhinitis was developed with Brown Norway rat and Sprague-Dawley rat, immunized with ovalbumin(OVA) emulsified in Al(OH)3 adjuvant. The nasal vascular permeability was assessed by determining the extent of the Evan’blue released into the nasal perfusate following the OVA challenge to the nasal cavity of rats. Results The release of Evan’blue following perfusion significantly increased in the sensitized BN rats and the released extent of Evan’blue was lower in the sham-sensitized BN rats. But there was no same result in SD rats. Conclusion The vascular permeability is accelerated by the immunized antigen of OVA in the sensitized BN rats and differs from that of sham-sensitized BN rats. Measurement of the extent of nasal vascular permeability in allergic rhinitis model can be considered as a useful experimental system for evaluating drug effects on allergic rhinitis. BN rat are more sufficient than SD rat in this experiment.
    Some Gender Difference on Stress Gastric Ulcer in Rats

    Objective To investigate the gender difference and potential mechanism of stress gastric ulcer in rats. Methods Fourty male and 40 female SD rats about 200-220 g were randomly divided into cold stress group (20 rats) and room temperature control group (20 rats) according to gender respectively.Rats of cold stress groups were afflicted gastric ulcer by cold stress method. Then the gastric ulcer index was compared between cold stress group of male and female groups, meanwhile the concentration of serum copper and zinc, activity of superoxide dismutase (SOD), and content of reduced glutathione (GSH) in every group were determined and analyzed. Results The gastric ulcer index in male group was significant higher than that in female group. Concentration of serum zinc upgraded and serum copper decreased after cold stress (P<0.05 or P<0.0l) and the serum copper of male group was more reduced(P<0.05). Activity of SOD in serum of both genders were decreased significantly (P<0.01), and the male group was even more reduced (P<0.01). Content of GSH in serum of both genders were increased overtly (P<0.01). Conclusion The rat’s gender have some effects on gastric ulcer of rats. Cold stress provokes copper and zinc re-distribution in vivo and free radical cleaning system differential priming response in two genders are part of the possible inherent mechanism of sexual difference in stress gastric ulcer.

    Objective To detect the activity of PLA2 and the content of PGE2 at local chronic compression injury in rat spinal cord. Methods Biochemical activity method was used to detect the activity of PLA2. Radio-immunity method was used to detect the content of PGE2 . Results (1) The activity of PLA2: The activity of PLA2 in experimental groups was increased after spinal cord compression, especial in the moderate compression group. There was significant difference (P<0.01) of the activity of PLA2 between the moderate compression group and the Sham-operated group.     Objective To explore the characteristics and role of pancreatic cell apoptosis in the acute phase of sodium taurocholate induced acute pancreatitis. Methods The electron microscopy was used for qualitative detection of apoptosis, terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) and flow cytometry were employed for quantitative detection of apoptosis. Results In pancreatitis rat model established, pancreatic tissuses were analysed. Electron microscopic detection: In the control group, ultrastructure of pancreatic acinar cells were normal. In the edema group, pathologic findings included formation of autophagic vacuoles and appearance of apoptotic cells and apoptotic bodies. In the necrotizing group, early apoptosis coexisted with necrosis and apoptosis was more significant than in the edematous group. TUNEL detection:At 3 hours, differences between the necrotizing and control groups were highly significant (P<0.01), those between the edematous and control groups were significant (P<0.05). There were no other significant differences among the groups. But apoptosis was the most significant in the necrotizing group.FCM detection: The results of FCM correlated positively with those of TUNEL. Conclusions Apoptosis is a kind of compensation, able to lighten the severity of the disease, but it can only act positively to a limited extent.

We systemically retrospected the methods, characteristic and development of myriads of metastatic animal models which have been established, and briefly introduced the application of new technique in the detection of micrometastasis formed in metastatic animal models.
